A new procedure has been developed for the rapid, large scale isolation of plasma membranes from Ehrlich ascites cells which is superior to other available methods. The surface charge density of these membranes (60,000 esu cm^-2) is 20 times the value reported for intact cells using different methodology. The Na+-K+-ATPase of these membranes has a low turnover number (2000 min^-1) and a high sensitivity to K+ at low [ATP]. Evidence is presented for the existence of an ATP-generating system in the membranes maintaining a steady state level of ATP of about 50 pmoles mg^-1.
