Novel roles for neurotrophins in the dopaminergic motor system

The neurotrophin brain-derived neurotrophic factor participates in cell survival, differentiation, maintenance and neuronal plasticity using both retrograde and anterograde axonal mechanisms. Dopaminergic neurons of the midbrain express high levels of BDNF, which is thought to be transported anterogradely to the striatum. To determine the role played by BDNF in the adult nigrostriatal system, we induced BDNF knockdown in the substantia nigra of adult mice using Cre/loxP site-specific recombination. Motor behavior was examined a few weeks post injection. A rotarod apparatus was used for motor coordination, an open field for spontaneous locomotor activity, a cylinder for laterality of voluntary forelimb use, a rotameter for spontaneous rotational asymmetry, grip strength was measured using a wire mesh grid, and clasping phenotype was quantified. Knockdown of BDNF gene expression in the substantia nigra induces significant abnormalities in spontaneous and rotational behavior. Results of clasping, rotarod, and grip strength are also abnormal. Our work suggests that BDNF expression in the nigrostriatal system, in addition to its known developmental survival role, has an important influence in maintenance of normal motor behavior in adult mammals. This novel anterograde neurotrophic support mechanism may also have important implications for mechanisms of motor dysfunction in Parkinson’s disease.La neurotrophine BDNF (brain derived neurotrophic factor) participe à la survie cellulaire, la différenciation, maintenance et la plasticité neuronale utilisant des mécanismes rétrogrades et antérogrades.
