Mutations in sodium/proton exchanger 9 (NHE9) and NHE6 have been associated with early developmental neurological disorders. These exchangers are thought to help regulate the luminal pH of vesicles within the endocytic pathway, a critical determinant of protein trafficking. We have previously shown that NHE6 localizes to recycling endosomes in neurons and undergoes activity-dependent redistribution in dendritic spines. Moreover, impaired NHE6 function results in reduced neuronal arborization and disrupts endocytic trafficking in developing neurons.
