Biotechnology of arsenic uptake and traslocation in plants (Solanum lycopersicum L.)

Il lavoro di ricerca di dottorato ha avuto come oggetto l'analisi a livello genetico e fenotipico della risposta di piante di pomodoro all'importante contaminante ambientale arsenico. La ricerca si è concentrata sulle interazioni genotipo-ambiente che portano varietà di pomodoro geneticamente diverse a reagire in modo differente alle stesse condizioni ambientali, con esperimenti in vitro e in serra che hanno coperto l'intero ciclo vitale delle piante. L'arsenico è stato studiato come contaminante inorganico presente in due forme chimiche, arseniato e arsenito, e i suoi effetti sono stati analizzati in una serie di varietà di pomodoro importanti per l'agricoltura italiana. Collaborazioni con aziende sperimentali e con James Hutton Institute (Regno Unito) hanno consentito di studiare nei dettagli il movimento delle forme di arsenico dal suolo ai tessuti delle piante, radici, foglie, steli e frutti. Una volta definite le diverse risposte rilevabili nelle varietà saggiate, si sono provate diverse pratiche agronomiche per migliorare le performance e minimizzare la contaminazione: aggiunta di silicati e impiego di biochar come ammendante. I risultati dimostrano innanzitutto che l’assunzione di arsenico nelle diverse forme e la sua traslocazione alle parti eduli sono molto diverse nelle diverse varietà. Analogamente, anche l'effetto dei silicati di riduzione della traslocazione è visibile solo in alcune cultivar, perché in altre si ottiene l'effetto contrario. L'aggiunta di biochar ha invece messo in luce gli effetti di questo ammendante sulla mobilità degli ioni nel suolo. Le ricerche sono state pubblicate in due articoli su riviste internazionali. La caratterizzazione della risposta all'arsenico è stata completata da analisi di espressione genica utilizzando geni candidati noti per essere coinvolti nella risposta delle piante a contaminanti inorganici.The research had the object of the analysis of the genetic and phenotypic response of tomato plants to arsenic contamination. The research has focused on genotype - environment interactions that lead to genetically different tomato varieties to react differently to the same environmental conditions, with in vitro and in vivo tests that covered the entire life cycle of plants. Arsenic has been studied as inorganic contaminants present in two chemical forms, arsenate and arsenite, and its effects have been analyzed in a number of tomato varieties important for the Italian agriculture. Experimental collaborations with different experimental farms and with the James Hutton Institute (United Kingdom) allows the study of the movement of the different arsenic species from soil to roots, leaves stems and fruits tissues. Once defined the different responses of the tested varieties, we try different agronomic practices to improve performance and minimize contamination: addition of silicates and use of biochar as a soil amendment. The results showed that the intake of arsenic in various chemical species and its translocation to the edible parts could be very different in the different varieties. Similarly, also the effect of reduction of the silicates on arsenic translocation is visible only in some cultivars, because in other cultivars it was observed an opposite effect. The addition of biochar has highlighted the effects of this fertilizer on the mobility of ions in the soil. The research has been published in two articles in international journals.
