A novel dendrimeric coating increases differentiation and enhances Wnt/b-catenin signaling in mesenchymal cells on rough titanium surfaces

Le strategie attuali per migliorare il successo clinico di impianti endo-ossei mirano a promuovere la guarigione e la formazione di tessuto osseo attorno agli impianti e quindi l’osteointegrazione. Per arrivare a questi obiettivi la ricerca si sta indirrizzando nella progettazione di superfici biomimetiche. L’idea nasce dal tentativo di replicare i stimoli biochimici che guidano le funzioni degli osteoblasti, mimando le composizioni strutturali e biochimiche dei tessuti biologici. Gli impianti endo-ossei, pero, devono severamente rispondere a dei requisiti meccanici in quanto la loro funzione impone di possedere proprieta meccaniche tali da sostenere le forze applicate nella sede d’impianto. Per migliare quindi la risposta biologica nella fase post-chirugica la ricerca si e mossa nella progettazione di rivestimenti superficiali implantari che possano condurre verso l’osteointegrazione. I polimeri ,grazie alla loro versatilita, risultano essere delle molecole promettenti per l’ideazione di tali rivestimenti. I dendrimeri sono polimeri ramificati che formano strutture 3D ben organizzate partendo da vari monomeri. La loro struttura puo essere disegnata finemente cambiando la composizione monomerica, creando cosi polimeri con proprieta chimico-fisiche e comportamenti biologici diversi. La loro plasticita le rende ideali per i bisogni della bioingegneria in quanto possono agire come un substratto abile a provedere ad un ancoraggio effettivo alle cellule comportandosi cosi come matrice extracellulare artificiale. La matrice extracellulare gioca un ruolo importante nello scambio di ossigeno e nutrimenti tra cellule e funge, inoltre, da reservoir per i fattori di crescita che possono essere rilasciati lentamente alle cellule circostatnti. Lo scopo di questo studio in vitro era quello di indagare il comportamento delle linee cellulari murine e cellule primarie stromali murine quando seminate su superfici di titanio diverse in presenza o assenza di rivestimenti innovativo dendrimerico di fosfo-serina/poli-lisina e osservare se quest’ultima puo influenzare positivamente la risposta cellulare. Risultati-I dendrimeri aumentano la proliferazione e adesione delle cellule MC3T3 nelle prime ore dopo la semina. Le cellule murine osteoblastiche MC3T3 sono state seminate su dischetti con superficie liscia (Polished), sand-blasted/acid atched (SAE) con e senza rivestimento e tenute in coltura in presenza di acido ascorbico per 6giorni. L’adesione cellulare, misurata mediante saggio MTT viene stimolata nelle superfici SAE+Co rispetto agli altri gruppi testati a 48 ore dalla semina. I dendrimeri aiutano il differenziamento delle linee cellulari osteoblasastiche murine. Per valutare gli effetti di questo rivestimento innovativo abbiamo scelto le cellule MC3T3-E1, cellule osteoblastiche murine isolate dalla calvaria di topi C57/BL6. Al sesto giorno di coltura l’espressione della fosfatasi alcalina si presentava sulla superficie SAE e ancora di più sulla SAE +Co significativamente più alta rispetto alla superficie liscia (Polished). Successivamente abbiamo misurato l’espressione di un diverso precoce marker osteoblastico, Osterix, il quale viene espresso nella fase pre-osteoblastica. I livelli trascrizionali sono risultati significativamente più alti sui dischetti SAE e SAE+Co rispetto ai dischetti Polished a 3 giorni. Questi valori si sono abbassati su tutte le superfici al sesto giorno rimanendo comunque, seppur di poco ma statisticamente significativo, più alto sulla superficie SAE+ Co. L’espressione di Osteocalcina (OCN) ha mostrato un trend diverso, infatti abbiamo notato un aumento dei livelli di mRNA di questo gene dal giorno 3 al 6 in tutti i gruppi testati. Nessuna differenza statisticamente significativa risultava al terzo giorno, ma le superfici con topografia ruvida presentavano livelli significativamente aumentati di Osteocalcina al giorno 6, inoltre la presenza di rivestimento dendrimerico ha accentuato questo aumento. Siccome la fosfatasi alcalina è un noto gene target della via canonica di Wnt abbiamo proceduto nel controllare se questa via è influenzata dalla presenza del rivestimento dendrimerico nelle cellule MC3T3. Si è analizzato l’espressione di Wisp-2 e Osteoprotegerina, geni sotto il controllo della via canonica di Wnt sulle superfici Polished, SAE e SAE+Co e si è notato un aumento dei livelli trascrizionali di questi geni sulla superficie SAE+Co dopo 6 giorni di coltura. Inoltre, i livelli mRna di β-catenina , uno dei fattori di trascrizione richiesti per questa cascata di segnalazione assieme al TCF, si presentano più alti sulla superficie SAE+Co a 6 giorni. Per poter misurare in modo diretto l’attività trascrizionale β-catenina/TCF abbiamo scelto un saggio reporter ed eseguito il test su cellule non differenziate murine C2C12 le quali presentano un modello vastamente utilizzato per lo studio di questa via. Come si nota dai grafici abbiamo un aumento della bioluminescenza sulle superfici SAE e SAE+Co. Presi nell’insieme questi dati mostrano come il rivestimento dendrimerico aumenta l’attivazione della via di Wnt e l’espressione dei geni target in cellule mesenchimali. I dendrimeri stimolano il differenziamento in cellule murine primarie In modo da poter confermare i risultati con delle linee cellulari primarie, abbiamo seminato cellule stromali midollari ossee prelevate da topi CD1 sui dischetti di titanio in presenza di acido ascorbico. In accordo con la letteratura ,abbiamo notato con l’aumento dell’espressione di Fosfatasi Alcalina e Osteocalcina che la topografia ruvida di superficie e la presenza del rivestimenti dendrimerico inducono l’espressione fenotipica osteoblastica più differenziata Lo sviluppo della moderna medicina è imprescindibile dal contemporaneo progresso tecnologico, che le mette a disposizione conoscenze, strumenti, materiali per poter raggiungere obiettivi di volta in volta più avanzati ed ambiziosi. Nell’ambito della biomimetica, il rivestimento dendrimerico testato può risultare di grande interesse nel migliorare le carattistiche dei biomateriali e favorire all’implantologia una ulteriore spinta verso un’osteointegrazione più celere.Current strategies to improve the clinical success of endosseous implant aim at promoting the biological pathways that sustain bone formation along the implant and thus osseointegration. To achieve this purpose biomimetic surfaces are being developed. Their rationale is to replicate the biochemical signaling governing osteoblast function, by mimicking the structural and biochemical composition of biologic tissues. Endosseous implants, however, have strict mechanical requirements, because their function requires them to possess mechanical properties enabling them to support significant mechanical loads, therefore there are unavoidable constraints in the material choice. This lead to the development of coatings, i.e. outer layers, surrounding a inner, generally titanium, core, possessing specific biological properties, and mimicking the bone tissue. Polymers are promising molecules for such coatings, because of their versatility. Dendrimers are branched polymers that form well organized 3D structures from various monomers. Their structure can be finely designed by changing the composition of the monomer, creating polymers with different physico-chemical properties and biological behavior. Therefore, their plasticity makes them suitable for the needs of bioengineering, because they can act as a substrate capable of providing an effective anchorage to cells, and as an artificial matrix to support the extracellular matrix (ECM) deposited by cells. The ECM plays a major role in defining cell behavior within a tissue and in contact of a biomaterial. The ECM provides the functional groups that can be bound by the cellular adhesion structures, it mediates the oxygen and nutrients exchanges between cells and blood vessels, it acts as a reservoir for growth factors, which can be thus slowly released to the surrounding cells, and of course it provide structural support to the tissue, transmitting the mechanical stimuli, which have been demonstrated to be central to the maintenance of bone homeostasis. Cells interact with the surrounding ECM through transmembrane proteins, the integrins, that are tightly connected to the internal cytoskeleton via a complex multi-molecular system regulating several aspects of cell fate. It has been demonstrated that adhesion can control the balance between cell survival and apoptosis (Gilmore, Owens et al. 2009), or that there is a strong association between cell shape and its commitment to a differentiation lineage . The goal of the present report was therefore to study the behavior of murine cell lines and primary cells from bone marrow, when cultured on commercially pure titanium surfaces in the absence or in the presence of a novel phospho-serine/poly-lysine biocoating, and investigate whether it could positively affect cell behavior. The murine osteoblastic cell line MC3T3 was plated on smooth (polished) titanium discs, sand-blasted, acid etched (SAE) titanium discs and SAE discs with biocoating (SAE+Co) and cultured in complete medium enriched with ascorbic acid for up to 6 days. Cell adhesion, as measured by MTT assay, was improved on SAE+Co when compared to the other groups at 48 hours. When we measured the expression of differentiation genes, we observed, from day 3 to day 6 of culture, an increase in the levels of transcripts of Alkaline Phosphatase and Osteocalcin, two markers of osteoblastic phenotype. This increase was higher in cells growing on SAE+Co surfaces. The expression levels of Osterix, an early osteoblastic marker decreased from day 3 to day 6 in all groups, but at day 6 they were higher on SAE+Co. Noticeably, the expression of Osteoprotegerin, a protein that antagonizes the formation of osteoclasts and a target gene of the canonical Wnt signaling, a fundamental pathway for the commitment to the osteoblast lineage, was higher in cells growing on SAE+Co both at 3 and 6 days of culture. Similarly, the mRNA levels of another target gene of the same cascade, Wisp-2 and of Ctnnb, the co-transcription factor mediating this pathway, were markedly increased in this group at day 6. To confirm the activation of Wnt/Ctnnb signaling pathway in cells growing on SAE+Co, we transfected the murine mesenchymal uncommitted cell line C2C12 on polished, SAE and SAE+Co surfaces with a reporter vector carrying the Firefly Luciferase gene under the control of a regulatory promoter sequence binding the dimer TCF/Ctnnb and a control vector expressing Renilla Luciferase in a constitutive way under the control of the CMV promoter. We then stimulated C2C12 with either vehicle or the soluble recombinant protein Wnt3a we observed an increase in the luciferase activity in all groups. This increase, however, was significantly higher in cells on SAE+Co. The results of the present study show that this novel biocoating improves cell adhesion and promotes the expression of a mature osteoblastic phenotype. Moreover, this biocoating stimulates the activation of Wnt canonical signaling, and enhances its activation by exogenous stimuli. Moreover dendrimers improve osteoblastic differentiation in murine primary bone marrow cells. - To confirm our results with an osteoblastic cell line, we cultured primary bone marrow stromal cells from CD1 mice in ascorbic acid for 10 days on Polished, SAE and SAE+Co discs. As several reports pointed out, rough surface topography induced the expression of a more differentiated osteoblastic phenotype, as shown by an increased expression of Alkaline Phosphatase and Osteocalcin. However, in accordance with our observations with MC3T3 cells, both ALP and OCN levels were markedly enhanced by the presence of the dendrimer biocoating. These result confirm that this coating promotes osteoblastic differentiation in a primary stromal cell model. The mechanisms through which dendrimers can enhance Wnt signaling however are still unknown. It can be speculated that dendrimers may be able to bind Wnt growth factors or other moieties and act as reservoir, further stimulating the downstream signaling. Alternatively, the stimulation of the Wnt pathway can be associated to the topographic features of the surface. We have shown that the coating can alter the surface profile in the nanometer range. The new profile might promote Wnt signaling to a greater extent
