On the basis of our results it can be concluded that the transfer of lipide-soluble CsA across the rat placental barrier in the materno-fetal direction is dependent on its maternal inflow concentrations. Moreover, an energy-dependent transport of CSA takes part during the passage of this substance across the rat placenta. When QND or CPZ are co-administered into the maternal reservoir together with CsA, the passage of CsA across the barrier is increased, which may suggest that QND and CPZ are able, probably by a competitive manner, to interact with a carrier for CsA. On the basis of pharmacokinetic examination, the following hypothesis has been created: P-gp returns CsA from the trophoblast cells back to the maternal compartment, and thus it restricts the transfer of CsA across the placental barrier.
