THE DIRECTED RECONSTRUCTION OF DNA. STUDYING THE STRUCTURE-FUNCTIONAL ORGANISATION OF alpha 2-INTERFERON OF THE MAN AND URIDINPHOSPHORILASE FROM ESCHERICHIA COLI

For the first time, the structure-functional investigation of field 30 - 50 a.o. of polypeptide chain alpha 2-interferon of the man has been performed, and the fragment, responsible for the antivirus activity of protein on the kines of cells of the man and of the pig, has been revealed. The model of the tertiary structure of the man's alpha 2-interferon antivirus site had been offered. The peculiarities of performance of the site-directed reconstruction of the DNA, using the uracylreparation system, have been investigated. The genes, coding the alpha D-interferon of the man and interferon- beta -Ser17, and strains-producers of the corresponding proteins, have been investigated. The new diagram of the solid-phase synthesis of aminooligonucleotides and non-radioactively marked probes on its base has been developed. Using the original modifications of the polymerase chain reaction method, the structure-functional investigation of uridinphosphorilase from Escherichia coli has been performed. Realized has been the synthesis of the eglin C gene (the inhibitor of proteinases from Hirudo medicinalis), and the strain-producer of eglin C has been constructed, which has created the prepositions for receiving the perspective preparation for the therapeutic application.
