Structure-function correlations of the reconstituted carriers have been derived from inhibition studies. Studying the reconstituted phosphate carrier from heart mitochondria new evidences for a common functional family of all mitochondrial carrier proteins were revealed. Catalytic functions of the identified amino acid units were discussed and compared with the single binding centre gate pore model. Results proved an asymmetric (righ-side out) orientation of the carrier in the proteolipsomes. The phosphate carriers at both sides of the membrane are inhibited by anions.
