Gentechnologische Bearbeitung des menschenpathogenen Hantavirus mit dem Ziel einen Impfstoff zu entwickeln Abschlussbericht

An increasing number of hanta virus infections has been reported recently. Serological methods (IFT, ELISA) are used to diagnose such infections in special laboratories. Genetically engineered hanta virus proteins are used for diagnoses and for vaccine development within the framework of this project. Virus-infected Vero-E6 cells and suitable sera from infected patients were introduced into indirect-IFT test kits. The baculo virus expression vector system (BEVS) and procaryotic vector systems were used for expression of recombinant hanta virus proteins. A commercial IFT kit was developed for detection of Nephropathia epidemica and the asiatic form of hemorrhagic fever with renal syndrome (HFRS). Both, the nucleocapsid proteins expressed in the eucaryotic BEVS and those expressed in the procaryotic systems are recognized by the sera of infected patients in the immunoblot and in the newly developed ELISA system. Work was started on the development of an IgM capture ELISA for detection of acute infections. Moreover, recombinant virus coat proteins are available for diagnostic uses and for vaccine development at a later stage.
