N-methyl-D-aspartátové (NMDA) receptory jsou ionotropní glutamátové receptory zapojené v excitačním synaptickém přenosu, synaptické plasticitě a excitotoxicitě. Jsou to heteromerní komplexy tvořené GluN1 spolu s GluN2A-D a/nebo GluN3A-B podjednotkami, které jsou aktivovány glutamátem a glycinem. Mnoho alosterických modulátorů může ovlivnit aktivitu těchto receptorů, včetně neurosteroidů. Pregnanolon sulfát (3α5βS) je endogenní neurosteroid, který inhibuje NMDA receptory use-dependentním způsobem a má neuroprotektivní účinek.
