Development of carbon sensor electrode for quantitative determination of urea

Doktora TeziDoktora Tezi olarak yapılan bu çalışmada, 4-NA olarak kısaltılan 4 nitroanilin molekülü kullanılarak camsı karbon (GC) elektrot yüzeyi dönüşümlü voltametri (CV) tekniği ile modifiye edilmiş, molekülün elektrokimyasal davranışları ve özellikleri aydınlatılmıştır. Çalışmanın esas amacı olan doğal numunelerde ürenin kantitatif tayinleri için ise 4-NA modifiye GC elektrot bir sensör elektrot olarak kullanılmış ve Kare Dalga Adsorptif Sıyırma Voltametrisi (SWAdsSV) tekniği kullanılarak ürenin süt numunesinde tayini yapılmıştır. Modifiye elektrotların yüzey karakterizasyonları, susuz ortamda asetonitrilde çözülmüş 100 mM NBu4BF4 destek elektroliti içerisinde hazırlanan 1.00 mM ferrosen (fcn) çözeltisi ile pozitif tarama yapılarak, sulu ortamda ise 10.0 mM H2SO4'de hazırlanmış 1.00 mM ferrisiyanür (HCF) çözeltisi ile negatif tarama yapılarak test edilmiştir. Ayrıca, yüzey karakterizasyon işlemlerinde 10.0 mM KCl içerisinde hazırlanan 1.00 mM Fe(CN)63-/4- redoks prob karışımı kullanılarak Elektrokimyasal İmpedans Spektroskopi (EIS) tekniğide kullanılmıştır. Çalışmada, SWAdsSV tekniği kullanılarak yapılan elektroanalitik ölçümler ile 1.00x10-12 M seviyelerine kadar inilmiştir. Ayrıca bu veriler ikinci bir metot olan HPLC tekniği ile kıyaslanmıştır. Metotlar arası uygunluğu test etmek için t ve F testleri yapılmış olup, istatistiksel olarak iki metot arasında bir fark bulunmamıştır.In this study, which was conducted as a doctoral thesis, by using 4 nitroaniline molecule, abbreviated as 4-NA, the glassy carbon (GC) electrode surface was modified with alternating voltammetry (CV) technique, and the electrochemical behavior and properties of the molecule were illuminated. For the quantitative determination of urea in natural samples, which is the main purpose of the study, the 4-NA modified GC electrode was used as a sensor electrode and the determination of urea in the milk sample was made using the Square Wave Adsorptive Stripping Voltammetry (SWAdsSV) technique. Surface characterizations of the modified electrodes were tested by positive screening with 1.00 mM ferrocene (fcn) solution prepared in 100 mM NBu4BF4 support electrolyte dissolved in acetonitrile in anhydrous environment, and negative scanning with 1.00 mM ferricyanide (HCF) solution prepared in 10.0 mM H2SO4 in aqueous environment. . In addition, Electrochemical Impedance Spectroscopy (EIS) technique was used in surface characterization processes using 1.00 mM Fe(CN)63-/4- redox probe mixture prepared in 10.0 mM KCl. In the study, electroanalytical measurements using the SWAdsSV technique were used to reach levels of 1.00x10-12 M. In addition, these data were compared with a second method, HPLC technique. T and F tests were carried out to test the compatibility between methods, and statistically there was no difference between the two methods
