La lyophilisation se divise en trois phases: la congélation, la dessiccation primaire et la dessiccation secondaire. Les différentes techniques d'optimisation de chaque phase visent toutes à réduire le temps de dessiccation, en permettant l'utilisation d'une température de dessiccation la plus importante possible. Celle-ci est limitée par la température de collapse du produit, qui est caractéristique d'une composition et des conditions opératoires. La définition de la température de collapse est donc primordiale. L'unique appareil qui permet de la mesurer est le microscope à platine de lyophilisation.
