Investigation of combined use of high intensity pulsed light, low voltage electrical treatment and natural antimicrobial substances for nonthermal pasteurization of clear apple juice

Bu tez kapsamında berrak elma suyunun ısıl olmayan yöntemlerle pastörizasyonu amacıyla yüksek yoğunluklu vurgulu ışık, düşük voltajlı elektriksel işlem, esansiyel yağ nanoemülsiyonu ilavesi ve bunların kombine etkilerinin meyve sularında önemli bir problem olan Alicyclobacillus acidoterrestris inaktivasyonu ve elma suyu kalite özellikleri üzerine etkileri incelenmiştir. Çalışmanın ilk aşamasında A. acidoterrestris üzerine antimikrobiyal etkisi olan esansiyel yağlar seçilmiş, bu esansiyel yağlar ve farklı emülgatörler kullanılarak nanoemülsiyonlar oluşturulmuş ve karakterize edilmiştir. Daha sonra stabilitesi yüksek olan nanoemülsiyonlar seçilerek elma suyuna ilave edilmiş ve duyusal açıdan kabul edilebilirliği yüksek olan nanoemülsiyon seçilerek çalışmanın devamında kullanılmıştır. Çalışmanın ikinci aşamasında, berrak meyve sularının ısıl olmayan pastörizasyonuna yönelik sürekli akış yüksek yoğunluklu vurgulu ışık (HIPL) sisteminin dizaynı, imalatı ve kurulumu gerçekleştirilmiştir. Ön denemeler yapılarak sistem test edildikten sonra, berrak elma sularına inoküle edilen A. acidoterrestris DSM 2498 vejetatif hücre ve sporlarının inaktivasyonu amacıyla farklı enerji dozlarında (0-12 J/cm2) HIPL uygulamasının etkisi incelenmiş, elde edilen inaktivasyon verileri modellenerek optimum uygulama dozu belirlenmiştir. Ayrıca, optimum noktada HIPL uygulaması süresince berrak meyve suyuna düşük voltajlı elektrik alan (5-30 V/cm) uygulamasının mikrobiyel inaktivasyon üzerine etkisi de incelenmiştir. Projenin son aşamasında ise Alicyclobacillus inaktivasyonu ve meyve suyu kalitesi açısından en iyi etkiyi gösteren bireysel ve kombine uygulamalar ile meyve suyu üretimleri gerçekleştirilmiştir. Aynı zamanda ticari olarak uygulanan geleneksel pastörizasyon ile berrak elma suyu örnekleri işlenerek kalite özellikleri ısıl olmayan teknolojiler ile işlenen örneklerle karşılaştırılmıştır. Elde edilen veriler ışığında; sürekli akış HIPL prosesinin berrak elma sularında A. acidoterrestris vejetatif hücre ve sporları üzerine inaktivasyon etkisinin daha fazla olduğu, fiziksel, kimyasal ve duyusal özelliklerinde olumsuz bir değişime neden olmadığı belirlenmiştir. Ayrıca, HIPL teknolojisi diğer ısıl olmayan teknolojilerle (düşük voltajlı elektriksel işlem ve esansiyel yağ nanoemülsiyonu ilavesi) kullanıldığında, geleneksel pastörizasyona alternatif olarak kullanılabileceği sonucuna varılmıştır.In this study, the effects of high intensity pulsed light, low voltage electrical treatment, and the addition of essential oil nanoemulsion on the pasteurization of clear fruit juices were studied. The effects of these methods on the inactivation of Alicyclobacillus acidoterrestris together with their effects on fruit juice quality were investigated. In the first part of the thesis, essential oils with a high antimicrobial effect on A.acidoterrestris were selected and nanoemulsions were formed by using these essential oils and different emulsifiers. After the characterization of nanoemulsions, nanoemulsions with high stability were added to apple juice for evaluating the sensory acceptability. Nanoemulsion with higher consumer acceptance was selected for further studies. In the second part, the continuous flow high intensity pulsed light (HIPL) system for non-thermal pasteurization of clear fruit juices was designed, manufactured and installed. Clear fruit juices were inoculated with A. acidoterrestris DSM 2498 vegetative cells and spores was exposed to HIPL with the different fluence (0-12 J/cm2). The obtained data was used for modelling the inactivation process and the optimum application dose was determined. The effect of low voltage electrical treatments (5-30 V/cm) on the microbial inactivation during HIPL were also investigated. In the last part of this thesis, fruit juices were produced by using the individual and combined processes showing the highest inactivation rate on A. acidoterrestris, and fruit juice quality was investigated during storage. The quality characteristics of clear apple juice produced by nonthermal technologies was compared to the commercially pasteurized apple juice sample. It was concluded that the continuous flow HIPL process was effective for the destruction of A. acidoterrestris vegetative cells and spores in clear apple juice without any adverse effect on physical, chemical and sensory properties. Furthermore, HIPL technology can be used as an alternative method to traditional pasteurisation when used with other non-thermal technologies such as low voltage electrical treatment and essential oil nanoemulsions
