Kabak ıslahında Fusarium dayanıklılığı üzerinde araştırmalar

The South-East of France provides 44% of the French national zucchini production mainly from the Gard area where localized in South of France around Nîmes. In summer 2019, after many years of monoculture production, a telluric disease causing wilting of the plant, crown necrosis with the browning of the plant vascular system was detected and impacted seriously the final yield of different zucchini fields. Suspecting that the pathogen belongs to the Fusarium genus, this master thesis has focused on the identification of the causal disease agent and the development of a pathogenicity test that could be used in a breeding program to select plant material with resistance against Fusarium spp. Diseased plants from Gard area were collected during two years and single spore culture were isolated and identified according to a morphological and molecular method. A total of 11 isolates collected from the field in Gard area, were identified as Fusarium solani f. sp. cucurbitae. In the greenhouse, three pathogenicity tests were conducted by using different methods of inoculation such as root dipping, pouring and direct inoculation of soil with pieces of agar covered with mycelium. The pathogenicity test using the root dipping method of inoculation was identified as the most relevant, repeatable and homogeneous test. The Fusarium solani f. sp. cucurbitae was determined as a very aggressive fungal disease agent killing Cucurbita pepo hybrids in only three weeks after inoculation. Although no resistant Cucurbits plants were detected during the pathogenicity test, some signs of resistance against disease agentwere identified in roots of C. maxima and C. moschata plant species which could be used in future Cucurbit breeding projectsFransa kabak üretiminin % 44'ü, Fransa'nın Güney-Doğusunda, Nîmes civarında bulunan Gard bölgesinden sağlamaktadır. 2019 yazında, uzun yıllar monokültür üretiminin ardından, toprak kaynaklı bitkinin solmasına ve bitkinin iletim sisteminde kararmaya neden olan bir patojen tespit edilmiş ve bu etmenin farklı kabak tarlalarında verimi önemli oranda azalttığı görülmüştür. Bu patojenin Fusarium cinsine ait olduğu şüphesi ile bu yüksek lisans tezinde, kabak üretilen arazilerde hastalık etmeninin tanımlanması ve Fusarium türlerine karşı dayanıklılık ıslahı programında kullanılabilecek patojenite testinin geliştirilmesi amaçlanmıştır. Gard bölgesinden hastalıklı bitkiler iki yıl boyunca toplanarak tek spor kültürü izole edilmiş ve bu izolatlar morfolojik ve moleküler olarak tanımlanmıştır. Gard bölgesindeki sahadan toplanan toplam 11 izolat, Fusarium solani f. sp. cucurbitae olarak tanımlanmıştır. Serada, kök daldırma, toprağa dökme ve miselyumla kaplı agar parçaları ile toprağı direk inoküle etme gibi üç farklı patojenite testi yapılmıştır. Kök daldırma testi, en iyi, tekrarlanabilir ve homojen test olarak belirlenmiştir. Fusarium solani f. sp. cucurbitae, inokulasyondan sadece üç hafta sonra Cucurbita pepo hibritlerini öldüren çok agresif fungal etmen olarak belirlenmiştir. Patojenite testi sırasında dirençli Cucurbita bitkisi tespit edilmemiş olmasına rağmen, C. maxima ve C. moschata bitki türlerinin köklerinde, gelecekteki Cucurbit ıslah projelerinde kullanılabilecek hastalık etkenine karşı bazı direnç belirtileri tespit edilmiştir
