Participación de las proneurotrofinas y sus receptores en la degeneración retiniana en modelos murinos de retinosis pigmentaria

219 pag.-63 fig.-6 tab.-anexos.[EN]Retinitis Pigmentosa (RP) is a term that encompasses a heterogeneous group of hereditary retinal dystrophy, for which there is currently no effective therapy. There are more than 300 mutations in about 60 RP-related genes; however, in most cases, they share a pathological and progressive death of photoreceptors that is directly related to vision loss and inexorably leads to blindness. For this reason, treatments, independent of the type of mutation underlying visual degeneration, but capable of protecting photoreceptors from damage and loss of function, even if they are only palliative remedies, are attractive therapeutic possibilities that are being actively attempted. Although several molecular mechanisms responsible for the death of photoreceptors in RP have been characterized, the order in which they act and their relative importance in the course of the disease are far from determined. However, results obtained in our group, as well as observations published by other laboratories, indicate the existence of a strong inflammatory component in this type of pathology, which allows to anticipate that damage in the retina is accelerated by imbalances in such inflammatory processes. Proneurotrophins are the precursor forms of neurotrophins. Although for years they were considered simple transition states in the maturation process of neurotrophins, they are currently known to be ligandos with their own activity that generally induce biological effects opposed to their mature forms, by binding to also different receptor complexes. Thus, while neurotrophins are involved in neuronal survival and differentiation, proneurotrophins generally trigger cell death processes. Signaling of the neural growth factor precursor (NGF), proNGF, through the p75 neurotrophin receptor (p75NTR), has been associated with different neurodegenerative disorders. Several models of retinal degeneration have a pro-inflammatory response mediated by proNGF. RP shares with these retinal pathologies numerous characteristics of the degenerative process, as well as the existence of a marked inflammatory component. This leads us to raise the possibility that the proNGF/p75NTR axis may also be involved in the progression of RP, contributing to the inflammatory imbalance. However, the participation of this proNGF/p75NTR axis in the progression of this disease has not been clearly described.Our work begins with the attempt to deepen knowledge of the cellular and molecular mechanisms involved in the inflammatory process that takes place in the retinas affected with RP. The results obtained have led us to explore the involvement of the proNGF-p75NTR axis in this pathology, as well as the neuroprotective potential of interfering with it, with the intention of defining new therapeutic targets. This study shows that, like microglia, GMCs have the ability to autonomously initiate a pro-inflammatory loop in dystrophic retinas. However, for the onset of gliotic response at the GFAP level, interaction between the two glia cell types is necessary. It also demonstrates the involvement of the proNGF/p75NTR axis in the progression of RP and the neuroprotective effect of interfering with it, in two different murine models. These results suggest that both, blocking the interaction between CGM and microglia, as well as blocking proNGF/p75NTR signaling, could result in independent of disease etiology therapeutic targets.[ES]La Retinosis Pigmentaria (RP) es un término que engloba un grupo heterogéneo de distrofias hereditarias de la retina, para las que actualmente no existe una terapia efectiva. Existen más de 300 mutaciones en unos 60 genes relacionadas con RP; sin embargo, la mayoría de los casos, comparten una muerte patológica y progresiva de los fotorreceptores que se relaciona directamente con la pérdida de visión y conduce inexorablemente a la ceguera. Por esta razón, los tratamientos, independientes del tipo de mutación que subyace a la degeneración visual, pero capaces de proteger a los fotorreceptores del daño y de la pérdida de función, aunque sean solo remedios paliativos, son posibilidades terapéuticas atractivas que se están intentando desarrollar activamente. Aunque se han caracterizado varios mecanismos moleculares responsables de la muerte de los fotorreceptores en RP, el orden en el que actúan y su importancia relativa en el curso de la enfermedad están lejos de haber sido determinados. No obstante, resultados obtenidos en nuestro grupo, así como observaciones publicadas por otros laboratorios, indican la existencia de un fuerte componente inflamatorio en este tipo de patologías, lo que permite anticipar que el daño en la retina se ve acelerado por desequilibrios en dichos procesos inflamatorios. Las proneurotrofinas son las formas precursoras de las neurotrofinas. Aunque durante años se consideraron simples estados de transición en el proceso madurativo de las neurotrofinas, actualmente se sabe que son ligandos con actividad propia que inducen, generalmente, efectos biológicos opuestos a sus formas maduras, mediante la unión a complejos receptores también distintos. Así, mientras que las neurotrofinas están implicadas en la supervivencia y diferenciación neuronal, las proneurotrofinas desencadenan, generalmente, procesos de muerte celular. La señalización del precursor del factor de crecimiento neural (NGF), el proNGF, a través del receptor de neurotrofinas p75 (p75NTR), ha sido asociada con diferentes trastornos neurodegenerativos. Diversos modelos de degeneración retiniana presentan una respuesta pro-inflamatoria mediada por el proNGF. La RP comparte con estas patologías de la retina numerosas características del proceso degenerativo, así como la existencia de un marcado componente inflamatorio.Esto nos lleva a plantear la posibilidad de que el eje proNGF/p75NTR esté implicado también en la progresión de la RP, contribuyendo al desequilibrio inflamatorio. Sin embargo, la participación del este eje proNGF/p75NTR en la progresión de esta enfermedad no ha sido descrita con claridad. Nuestro trabajo se inicia con el intento de profundizar en el conocimiento de los mecanismos celulares y moleculares implicados en el proceso inflamatorio que tiene lugar en las retinas afectadas con RP. Los resultados obtenidos nos han llevado a explorar la implicación del eje proNGF-p75NTR en esta patología, así como el potencial neuroprotector de interferir en el mismo, con la intención de definir nuevas dianas terapéuticas. Este estudio demuestra que al igual que la microglía, las CGM tienen la capacidad para iniciar un circuito proinflamatorio en las retina distróficas de manera autónoma. Sin embargo, para el inicio de la respuesta gliótica a nivel de GFAP, es necesaria la interacción entre ambos tipos celulares. Así mismo, se demuestra la implicación del eje proNGF/p75NTR en la progresión de RP y el efecto neuroprotector de interferir en el mismo, en dos modelos murinos distintos. Estos resultados sugieren que, tanto el bloqueo de la interacción entre CGM y microglía, como el bloqueo de la señalización proNGF/p75NTR podrían suponer dianas terapéuticas independientes a la etiología de la enfermedad.Beca FPIPeer reviewe
