Optimization of small scale lipid extraction method for freshwater fish (Barbodes banksi) and marine fish (Polydactylus sextarius) muscles are conducted based on Folch (1957) lipid extraction procedure. The optimization process was conducted by comparing different homogenization methods, extraction solvent ratios, and sample weight to solvent ratios to achieved high percentage of lipid recovery. On the first optimization, three methods of tissue homogenization (glass beads, Mortar and pestle, and mincing by dissecting blade) were done and glass beads was found to be optimal method for both freshwater fish and marine fish muscle tissue.
