Recent studies have shown that a cellulolytic bacterium, Clostridium thermocellum, can be genetically modified. However, strong expression of heterologous genes and secretion of their proteins in this organism are difficult. In the current study, we report that a gene expression cassette including the cellobiose phosphorylase gene promoter, the hypoxanthine phosphoribosyl transferase gene terminator from C. thermocellum, and the ribosome-binding site and histidine-tag sequence from pQE30, led to the expression of a heterologous gene: aldehyde deformylating oxygenase.
