目的研究姜黃素對NB4細胞組蛋白H3和非組蛋白P53的乙酰化和細胞增殖的作用,探討姜黃素抗白血病機制。方法應用不同濃度(50,25,12.5,6.25,3.125μmol/L)的姜黃素作用NB4細胞不同時間(0,4,8,12,24h),MTT法測定姜黃素對NB4細胞增殖的影響,Western blot法檢測乙酰化組蛋白H3和乙酰化P53的水平。結果姜黃素以時間和劑量依賴方式抑制NB4細胞增殖,在24h和36h的IC50值分別為40μmol/L和25μmol/L;能明顯上調組蛋白H3的乙酰化水平,促進P53的表達和P53的乙酰化。結論姜黃素具有去乙酰化酶抑制劑作用,能上調組蛋白H3乙酰化水平,促進腫瘤抑制因子P53表達和活化,抑制白血病細胞增殖。OBJECTIVE: To investigate the effects of curcumin on the acetylation of histone H3, P53 and the proliferation of NB4 cells.
