Creation of Polyploid by Colchicine Treatment on Seedlings within Tube of Robinia pseudoacacia L.

In the polyploidization of chromosome by colchicine treatment on seedlings, there was various difficult points. Namely, the ratio of the polyploidization is generally very low and the inducement of polyploid is localized into only a few joints. And so, we tested the use of tissue culture technique to overcome all difficulties in relation to induce of polyploid. The tetraploid plantlets of Niseakasia (Robinia Pseudoacacia L.) were induced effectively by culture of callus originated from seedlings treated with colchicine.コルヒチン処理による染色体の倍数化は, コルヒチン処理による倍数化率がきわめて低く, また局部的変異に限定されるという難点があった. そこで, このような難点を克服するために組織培養の利用を試みた. その結果, コルヒチン処理をニセアカシアの無菌の芽生えに行い, その各組織からカルスを誘導し, カルス培養を行ったところ4倍体の幼植物体を効果的に再生することができた
