Comparative analysis of derived neurons from induced pluripotent stem cells (iPSC) to study the Gaucher disease and Parkinson\'s disease association

A doença de Parkinson (PD) (PD - Parkinson Disease, MIM # 168600) é uma doença neurodegenerativa caracterizada por morte de neurônios dopaminérgicos da parte compacta da substancia negra do mesencéfalo (mDAN). Pacientes com PD exibem frequência aumentada de mutações no gene GBA, que codifica a enzima beta-glicosidase ácida (ou glicocerebrosidase - GCase) deficiente na Doença de Gaucher (GD), indicando uma forte associação entre as duas patologias. Atualmente não se conhece o mecanismo pelo qual mutações em GBA aumentam a susceptibilidade à degeneração neuronal. O conhecimento dos mecanismos moleculares desta associação contribuirá para uma maior compreensão das bases fisiopatológicas da neurodegeneração na PD permitindo o desenvolvimento de novas estratégias terapêuticas. Nos últimos anos, nosso grupo identificou pacientes com PD portadores de mutações no gene GBA e coletou amostras de sangue de 3 diferentes grupos fenotípicos: pacientes com PD sem mutação no gene GBA (PD), pacientes com PD e mutação no gene GBA (PDGBA: 2 heterozigotos e 1 homozigoto recessivo apresentando GD) e controles sadios (Ctrl). Aqui, descrevemos o uso de hiPSCs derivadas destes pacientes para a análise do papel de GBA na PD. As linhagens foram diferenciadas em neurônios dopaminérgicos A9 do mesencéfalo (iDA), principal tipo celular acometido pela PD, em 3 condições de cultivo: normóxia, hipóxia e em normóxia sob estímulo de campo magnético externo (EMF). As análises de produção de espécies reativas de oxigênio (ROS) revelou diminuição da produção de ROS e de atividade lisossômica em células de pacientes PD com mutação em GBA quando comparados com células de controles e de pacientes PD em hipóxia. Os resultados de análise de RNAseq indicam que a assinatura gênica para neurônios mDAN maduros foi maior na condição de hipóxia. Também foram observadas diferenças significativas no perfil de expressão gênica relacionados a diferentes vias metabólicas, revelando um papel exacerbado de respostas inflamatórias em pacientes PD com mutações em GBA. Propomos que a deficiência de atividade lisossômica causada por GBA cria um ciclo de inflamação exacerbando desse fenômeno já descrito na PDParkinson\'s disease (PD) (PD - Parkinson Disease, MIM # 168600) is a neurodegenerative disease characterized by the death of dopaminergic neurons from substantia nigra pars compacta in the midbrain (mDAN). PD patients exhibit increased frequency of mutations in the GBA gene, which encodes the enzyme glucocerebrosidase (GCase) deficient in Gaucher Disease (GD), indicating a strong association between the two pathologies. Currently, the mechanism by which GBA mutations increase neuronal degeneration susceptibility is unknown. Knowledge of the molecular mechanisms of this association will contribute to a better understanding of the pathophysiological bases of neurodegeneration in PD, allowing the development of new therapeutic strategies. In the last years, our group has identified PD patients with mutations in GBA gene and collected blood samples from 3 different phenotypic groups: PD patients without a GBA gene mutation (PD), PD patients plus GBA gene mutation (PDGBA: 2 heterozygotes and 1 recessive homozygote presenting GD) and healthy controls (Ctrl). Here, we describe the use of hiPSCs derived from these patients to analyze the role of GBA in PD. The cell lineages were differentiated into dopaminergic neurons A9 of the midbrain (iDA), the main cell type affected by PD, in 3 culture conditions: normoxia, hypoxia and in normoxia under external magnetic field (EMF) stimulation. The production analysis of reactive oxygen species (ROS) revealed a decrease in ROS production and lysosomal activity in cells of PD patients with GBA mutation when compared to control cells and PD patients in hypoxia. The results of RNAseq analysis indicate that the gene signature for mature mDAN neurons was greater in the condition of hypoxia. Significant differences were also observed in the gene expression profile related to different metabolic pathways, revealing an exacerbated role of inflammatory responses in PD patients with GBA mutations.
