The hepatitis C Virus (HCV) RNA-dependent RNA-polimerase (NS5B) shows a great genetic diversity reflected in the high number of circulating genotypes and subtypes. This polymerase initiates RNA synthesis by a de novo mechanism. This process implies conformational changes that direct transition from the initiating complex to a processive elongation complex. Most of the studies with the NS5B polymerase have been done with genotypes 1b and 2a, while information about other genotypes is scarce.
