In der vorliegenden Arbeit werden die positiven Effekte des Phosphodiesterase-5-Inhibitors Sildenafil auf die Gefäßfunktion in einem experimentellen Diabetes-Modell (Streptozotocin-induziert) beschrieben. Diabetes und seine kardiovaskulären Folgeerkrankungen, welche bis zu 80 % der Todesfälle bei diabetischen Patienten verursachen, gehen mit einer Endotheldysfunktion einher, bei der eine verminderte NO-Bioverfügbarkeit sowie eine verstärkte Sauerstoffradikalbildung eine Schlüsselrolle spielen. Endotheliales NO aktiviert die lösliche Guanylatzyklase in glatten Gefäßmuskelzellen, welche über erhöhte zyklische Guanosinmonophosphat-Konzentrationen eine Gefäßrelaxation bewirkt.
