Involvement of the Wnt/β-catenin signaling pathway in the primary and metastatic tumor development of osteosarcoma

L'ostéosarcome est la tumeur osseuse primitive maligne la plus fréquente chez les enfants avec 150 nouveaux cas par an en France. La survie des patients n’a pas évolué depuis 30 ans, atteignant 70 % à 5 ans pour les patients présentant une forme localisée de la maladie, mais seulement 25 % en présence de métastases (20% des cas), aucun traitement ne montrant une efficacité suffisante chez ces patients métastatiques. Il est donc nécessaire de développer de nouvelles stratégies thérapeutiques. Dans ce contexte, une dérégulation de la voie de signalisation Wnt/β-caténine a été rapportée dans de nombreux cas d'ostéosarcome, mais son implication dans le développement de ces tumeurs reste controversée. Ainsi, nous avons évalué le potentiel anti-tumoral de l'ICG-001, une molécule ciblant spécifiquement l’interaction entre la protéine de liaison CREB Binding Protein (CBP), et la β- caténine et inhibant ainsi la transcription dépendante de ce complexe, dans trois lignées cellulaires d’ostéosarcome humain, KHOS, MG63 et SJSA1. L'ICG-001 diminue la prolifération cellulaire mais, de façon surprenante, favorise la migration des cellules in vitro et le développement de métastases pulmonaires dans un modèle murin de xénogreffe induit par une injection para-tibiale de cellules KHOS. Cet effet pro-migratoire pourrait résulter en partie, d’un déplacement d’équilibre transcriptionnel d’un complexe se composant de la β-caténine et de CBP vers un complexe incluant la β-caténine et un autre cofacteur p300. Cette étude ajoute donc un niveau de complexité quant au rôle de la voie Wnt/β-caténine dans le développement métastatique de l'ostéosarcome.Osteosarcoma is the most common primary malignant bone tumor in children with 150 new cases per year in France. Patient survival has not been improved for the last decades, reaching 70% to 5 years for patients with localized disease, but only 25% for patients with metastatic disease (20% of cases), no effective treatment could develop for these patients with metastases at diagnosis. Therefore, it is necessary to develop new therapeutic strategies. In this context, a deregulation of the Wnt/β-catenin signaling pathway has been reported in many cases of osteosarcomas, but its involvement in the development of these tumors remains controversial. Thus, we evaluated the anti-tumor potential of ICG-001, a small molecule specifically targeting the interaction between the CREB Binding Protein (CBP) and the β- catenin, inhibiting the dependent transcription of this complex, in three human osteosarcoma cell lines, KHOS, MG63 and SJSA1. ICG-001 reduces cell proliferation but, surprisingly, promotes cell migration in vitro and the development of pulmonary metastases in a mouse xenograft model induced by para-tibial injection of KHOS cells. This pro-migratory effect could result in part from a transcriptional switch from a complex consisting of β-catenin and CBP to a complex including β-catenin and another cofactor, p300. Thus, this study adds a level of complexity to the role of the Wnt/β- catenin pathway in the metastatic development of osteosarcoma
