Analysis of model of tryptophan metabolism

Matematičko modeliranje daje nam na jednostavan i ponovljiv način detaljan uvid na metaboličke procese koji se zbivaju u stanicama. Metabolizam triptofana uglavnom se odvija preko kinureninskoga puta pri kojem nastaju metaboliti zajedničkoga naziva kinurenini. U ovom radu provedena je analiza modela metabolizma triptofana programima CellDesigner i WR Mathematica. Primjenom analize parametarske osjetljivosti identificirane su najosjetljivije reakcije modela. Rezultati provedene analize pokazuju da su reakcija sinteze proteina i reakcije transporta triptofana između cirkulacije i pojedinih tkiva najosjetljivije reakcije u modelu. Enzim aril formamidaza (AFD) uključen u kinureninski put pokazao se kao ključni enzim za analizirani model metabolizma. Ovaj model uspješno simulira metabolizam triptofana kinureninskim putem te se može koristiti za buduća eksperimentalna istraživanja.Mathematical modeling gives us a simple and reproducible detailed insight into the metabolic processes that take place in cells. Tryptophan metabolism is mainly happening via the kynurenin pathway, which produces metabolites collectively known as kynurenines. In this paper the analysis of tryptophan metabolism model was performed by CellDesigner and WR Mathematica. By applying parametric sensitivity analysis, the most sensitive reactions of the model were identified. The results of the analysis show that the protein synthesis reaction and the tryptophan transport between the circulation and individual tissues are the most sensitive reactions in the model. The enzyme aryl formamidase (AFD) involved in the kynurenine pathway has been shown to be a key enzyme for the analyzed metabolism model. This model successfully simulates tryptophan metabolism by the kynurenin pathway and can be used for future experimental research
