Thrombin activatable fibrinolysis inhibitor (TAFI) and plasminogen activator inhibitor-1 (PAI-1) are important regulators of fibrinolysis. Bothproteins attenuate the tissue-type plasminogen activator (tPA)-mediatedconversion of plasminogen into fibrin-degrading plasmin, however with distinct modes of action. Activated TAFI (TAFIa) removes C-terminal lysines from the fibrin surface, rendering fibrin less susceptible to fibrinolysis.
