Immune response and conditional role of pathogenic bacteria in the pathogenesis of perimucositis and periodontitis in the elderly

Periodontitis and periimplantitis are the two major oral diseases encountered in the practice of periodontal tissue therapy. These are polymicrobial inflammatory diseases that disrupt the structures supporting the tooth or implant and result is the loss of the tooth / implant without treatment. Cytokines form a group of mediators of nonspecific inflammatory responses, are soluble, low molecular weight proteins that, under physiological and pathological conditions, transmit certain intercellular signals required for an integrated cellular response to different external stimuli. Objective of the work: to research the role of immune response and conditionally pathogenic bacteria in the peri-implant tissue after treatment with implants and dental prosthesis in elderly patients with periodontitis or edentulous patients with peri-mucositis. The tasks: 1. To determine and evaluate changes in interleukin 1β, IL-10 concentration in the medium of stimulated, unstimulated and immunomodulatory-treated peripheral blood leukocytes, in the peri-mucositis group and the group with healthy tissue around implants. 2. To determine and evaluate changes in local inflammatory markers IL-17, TNF-α, and collagenase MMP-8 activity in tissue fluid around the dental implant in peri-mucositis group and in healthy subject group. 3. To determine the peculiarities of pathogenic bacteria in the pockets of dental implants in peri-mucositis group and in healthy subject group, and to establish correlations with markers of local inflammation. 4. To determine and evaluate changes in interleukin 1β concentration in the medium of stimulated and unstimulated peripheral blood leukocytes in the periodontitis group and in the group with healthy tissue around implants and to investigate the influence of Staphylococcus aureus and Escherichia coli on leukocyte secretion by this mediator
