Mechanisms of free hemoglobin binding and removal from blood plasma.

Wolna hemoglobina, występująca poza erytrocytami, zarówno z przyczyn fizjologicznych, jak i patologicznych (choroby związane z hemolizą), wymaga istnienia szeregu mechanizmów, wychwytujących i eliminujących tę cząsteczkę z osocza, aby nie dopuścić do jej szkodliwego działania na organizm. Po wydostaniu się z erytrocytów, hemoglobina jest silnie wiązana przez białko ostrej fazy haptoglobinę, z którą tworzy trwały kompleks. Wolny hem, po związaniu przez białko hemopeksynę, jest usuwany z osocza krwi za pośrednictwem receptora LRP/CD91. Układ wychwytu i eliminacji z osocza wolnej hemoglobiny, jak również jej kompleksu z haptoglobiną, stanowi receptor CD163, należący do grupy receptorów zmiatających, występujący przede wszystkim na powierzchni makrofagów i monocytów, oraz receptory megaliny i kubiliny znajdujące się w kanalikach bliższych nerek.Free hemoglobin, when present outside erythrocytes, for either physiological or pathological causes (diseases associated with hemolysis) can have adverse effects on the organism. Therefore it requires a number of mechanisms that can bind and eliminate this molecule from the plasma to prevent such effects. Hemoglobin released from erythrocytes, is strongly bound by an acute phase protein, haptoglobin, and forms with it a stable complex. This complex, can then bind to CD163 receptor, belonging to the scavenger family receptors, found mostly on the surface of macrophages and monocytes. Also free heme, after binding to another protein hemopexin is removed from the plasma via LPR/CD91 receptor present mostly on macrophages. The system of uptake and elimination from the plasma of free hemoglobin includes also receptors megalin and kubilin, which are located in cells lining proximal renal tubules
