Acute experimental peritoneal inflammation induced by Saccharomyces cerevisiae antigens - a role of eicosanoids in vascular permeability

Inflammation is the body's reaction to the damage of cells/tissues or infection with pathogenic microorganisms. Damaged cells release signals to inform the immune system of a danger, and the released mediators are recognized by cell receptors and stimulate the production of proinflammatory mediators. Inflammation consists of several steps, the first of these is to identify the pathogen, another is to increase the vascular permeability, and it is followed by the influx of leukocytes into the inflammatory focus. Leukocytes that reach the site of infection eliminate the pathogen, and subsequently the resolution of inflammation is initiated.The aim of the thesis was to characterize the process of increased vascular permeability during acute experimental peritoneal inflammation induced by antigens of Saccharomyces cerevisiae (zymosan) and clarify the role of eicosanoids in the process.The role of eicosanoids in vascular permeability in the above model was analyzed and described by several research groups, from the pioneering studies by Doherty et al. (Prostaglandins, 1985, 30:769) through investigations of Kolaczkowska et al. (Inflammation, 2002, 26) and Maekawa et al. (J Biol Chem 2002, 277:20820). These studies have shown that eicosanoids, especially prostaglandins (PGE2, PGI2) and cysteinyl leukotrienes (LTC4-LTE4), are largely responsible for the increased permeability of blood vessels during zymosan peritonitis. Most importantly, the above reports demonstrated that resident peritoneal leukocytes, mast cells and particularly macrophages, are important cellular sources of eicosanoids and the resident leukocytes are responsible for vascular changes.Summarizing, we conclude that studies on the model of peritonitis induced by zymosan lead to understanding of general mechanisms associated with the changes of vascular permeability accompanying acute inflammation.Zapalenie jest reakcją organizmu na uszkodzenie komórek/tkanek lub zakażenie mikroorganizmami chorobotwórczymi. Uszkodzone komórki uwalniają sygnały informujące układ immunologiczny o niebezpieczeństwie, a niektóre z tych mediatorów są rozpoznawane przez receptory komórkowe stymulujące wytwarzanie mediatorów prozapalnych. Zapalenie składa się z kilku podstawowych etapów, pierwszym z nich jest rozpoznanie patogenu, kolejny to wzrost przepuszczalności naczyniowej, po którym następuje napływ leukocytów do ogniska zapalnego. Jeżeli leukocyty dostaną się do miejsca infekcji następuje eliminacja patogenu, a po usunięciu czynnika chorobotwórczego zachodzi ostatni etap, którym jest wyciszenie zapalenia. Celem pracy była charakterystyka procesu zwiększonej przepuszczalności naczyniowej w trakcie ostrego eksperymentalnego zapalenia jamy otrzewnej wywołanego antygenami Saccharomyces cerevisiae (zymosan) i wyjaśnienie roli eikozanoidów w tym procesie.Rolę eikozanoidów w przepuszczalności naczyniowej na powyższym modelu analizowało i opisało kilka grup badawczych, począwszy od pionierskich badań Dohertiego et al. (Prostaglandins, 1985, 30:769) oraz Kołaczkowskiej et al. (Inflammation, 2002, 26) i Maekawy et al. (J Biol Chem, 2002, 277:20820). Powyższe badania dowiodły, że eikozanoidy, a zwłaszcza prostaglandyny (PGE2, PGI2) i leukotrieny cysteinowe (LTC4-LTE4) są w znacznym stopniu odpowiedzialne za zwiększenie przepuszczalności naczyń krwionośnych podczas ostrego zapalenia otrzewnej. Co więcej wykazano, że spośród leukocytów otrzewnowych mastocyty, a zwłaszcza makrofagi, są ważnymi źródłami komórkowymi eikozanoidów i leukocytami odpowiedzialnymi za zmiany naczyniowe. Podsumowując można stwierdzić, że badania na modelu zapalenia otrzewnej wywołanego przez zymosan umożliwiły poznanie mechanizmów związanych ze zmianami naczyniowymi towarzyszącymi zapaleniu
