International audienceThe advent of designer nucleases, especially CRISPR-Cas9, is providing a real breakthrough in genomeediting technology. The principle is simple : Double-strand breaks (DSB) are accurately produced onselected target DNA sequences in specific cells. These DSB are then repaired by non-homologousend-joining, resulting in inactivation of the targeted gene(s), or by inserting a specific DNA fragment (template) through homologous recombination. These technologies can be used in multipledomains, including fundamental research, gene therapy, ecological engineering, biotechnologies oragriculture.
