Ph.D.The CRISPR/Cas9 technology has demonstrated great potentials in genome editing with its superior efficiency and flexibility and it has been widely applied for various studies. To explore its capacity in delineating sophisticated gene functions during cellular differentiation, we initiated this project with two parts of works.In the first part, we developed an inducible Ert2-Cas9 system to render controllable genome editing. We showed that the Ert2-Cas9 showed functional activity in the presence of tamoxifen. Based on similar design, we established the Ert2-Cpf1 system. Further, we introduced Cre/LoxP system as a secondary controller to overcome the Ert2 leakage problem and enhance the inducible feature, termed as TD-Ert2-Cas9. By this ...
