It is well known that the double-stranded RNA-dependent Protein Kinase (PKR) is not only activated in various models of Alzheimer's disease (AD) and in AD patients but also associated with neurodegeneration and cognitive decline observed in this dementia. Recent studies reported that PKR could be involved in the control of inflammatory factors expression, including (TNFalpha). This study aims to investigate the effects of the PKR pharmacological inhibition on the production and the release of TNFalpha in a mouse Amyloid Beta (Abeta42) -treated co-culture containing physiological proportions of neurons (45%), astrocytes (40%) and microglia (15%), and also to test the specific PKR inhibitor on human lymphocytes of AD patients.
