NIR-spectrometry as a method of quantitative analysis of synthetic and phytogenic agents in tablets and granulates

The NIR-spectrometric measurement of the diffuse reflectance is a powerful tool for the quantitative multicomponent analysis of solid products. In the past it could be shown that this works well with powdery samples. Pulverizing of finished products however may be troublesome and often lacks reproducibility. In this study it was tried to perform a quantitative analysis by measuring directly the reflection of a Tolbutamid tablet. Further it was tried to perform a quantitative analysis by measuring directly the reflection of an active agent in a phytogenic matrix (Drug). The active substances studied in this paper are the 1,8-dihydroxyanthrone derivates which due to their origin from the Senna Plant are called sennosides. This however requires a previous calibration with standards the concentrations of which have to be determined by an adequate reference method. Die NIR-spektrometrische Messung in diffuser Reflexion ist ein leistungsfähiges Werkzeug für die quantitative Mehrkomponentenanalyse von Feststoffen. In der Vergangenheit wurde bei entsprechenden Analysemethoden meist vorausgesetzt, daß die Proben pulverförmig vorliegen. Die Pulverisierung von Fertigprodukten ist jedoch relativ umständlich und häufig nicht ausreichend reproduzierbar. In dieser Arbeit wird eine quantitative Wirkstoffanalyse durch direkte Reflexionsmessung an Tolbutamidtabletten durchgeführt. Ferner wird versucht, eine quantitative Wirkstoffanalyse durch direkte Reflexionsmessung von pharmazeutischen Wirkstoffen in einer pflanzlichen Matrix (Droge) durchzuführen. Die hier untersuchten Wirkstoffe sind 1,8-Dihydroxyanthron-Derivate, die nach ihrer Herkunft, der Senna-Pflanze, als Sennoside bezeichnet. Um eine entsprechende NIR-Kalibration erstellen zu können, benötigt man Standards, deren Gehalt mit einer geeigneten Referenzmethode ermittelt werden muß
