Determination of population structure by using genotyping by sequencing (GBS) in common bean (Phaseolus vulgaris L.)

Bu çalışmada, Türkiye ve farklı ülkelerden toplanmış, 173 adet Phaseolus vulgaris genotipleri arasındaki genetik çeşitliliğin ve popülasyon yapısının Genotyping by Sequencing (GBS) metodu ile belirlenmesi amaçlanmıştır. Bu çalışma ile ilk kez fasulyede GBS metodu kullanılmıştır. GBS metodu ile fasulye genomunda bulunan Single Nucleotide Polymorphism (SNP) markırları saptanmıştır. GBS analizi sonucunda 1500 SNP elde edilmiştir. Markırların ortalama PIC değeri 0,2 olarak tespit edilmiştir. SNP markırları kullanılarak STRUCTURE programında popülasyon yapısı belirlenmiştir ve 173 genotip kendi arasında 3 guruba (K=3) ayrılmıştır. Elde edilen bulgulara göre Andean ve Mezoamerikan gruplarına başarılı bir şekilde ayrıldığı görülmüştür. Fasulyede ilk kez yüksek yoğunluklu markır kullanılarak genetik çeşitlilik analizi gerçekleştirilmiştir.The aim of this study was to determine genetic diversity among genotypes and population structure by using Genotyping by Sequencing (GBS) in common bean (Phaseolus Vulgaris L.) . In this study, x173 different genotypes from different regions of World were used. This is the first usage of GBS in common bean. We scored 1500 SNP's by using GBS method. Population structure was determined using STRUCTURE software and mainly three groups (K=3) were identified among 173 common bean genotypes. PIC value was determined as 0,2 . Overall Andean and Mezoamerican gene pools were succesfuly distinguish the population structure of common bean
