Production and purification of levansucrase enzyme by Zymomonas mobilis

Levansukrazlar (E.C.2.4.1.10), GH68 glikozit hidrolaz enzim ailesine ait olup sakkarozu substrat olarak kullanarak levan ve levan tipi fruktooligosakkaritlerin (FOS) oluşumunu katalizleyen fruktoziltransferaz grubu enzimlerdir. Bu çalışmada Zymomonas mobilis subsp. mobilis NRRL B-14023 ile levansukraz üretimi üzerine fermantasyon süresi, başlangıç substrat konsantrasyonu, sıcaklık ve pH'nın etkileri incelenmiş ve bunlardan; sıcaklık (28-32 °C) başlangıç sakkaroz konsantrasyonu (100-200 g/L) ve pH'nın (4.5-5.5) levansukraz üretimine etkilerini incelemek ve bu parametrelerin optimum seviyelerini belirlemek amacıyla Cevap Yüzey Yöntemi (CYY) kullanılmıştır. Başlangıç substrat konsantrasyonu, sıcaklık ve pH'nın levansukraz üretimi üzerine önemli bir etkiye sahip olduğu, regresyon modelinin önemli (P0.05) olduğu belirlenmiştir. Deneysel veriler kullanılarak bir matematik model oluşturulmuş ve bu verilerle modelden elde edilen verilerin uyumlu olduğu tespit edilmiştir. Bu model yardımıyla proses için optimum koşullar olan 159.01 g/L başlangıç substrat konsantrasyonunda, 30.3 °C sıcaklıkta ve pH 4.91'de modelin tahminlediği aktivite değeri 13.18 mM glukoz/dak iken, üretim sonunda 13.3 mM glukoz/dak levansukraz aktivitesi elde edilmiştir. Zymomonas mobilis NRRL B-14023 ile üretilen levansukraz enzimi kullanılarak FOS üretimi gerçekleştirilmiştir. Ayrıca Cevap Yüzey Yöntemi ile belirlenen optimum koşullarda üretilen enzim saflaştırılmış ve SDS-PAGE ile saflaştırılan enzimin molekül ağırlığı yaklaşık 48 kDa olarak belirlenmiştir.Levansucrases (E.C.2. 4. 1. 10) are fructosyltransferase (Ftase) which belong to GH68 glycoside hydrolase enzyme family and catalyze the formation of levan and fructooligosaccharides (FOS) using sucrose as substrate. In this study, the effects of initial substrate concentration, fermentation time, temperature and pH on levansucrase production by Z. mobilis subsp. mobilis NRRL B-14023 was investigated. Response Surface Methodology was used optimized the enzyme production based on three selected parameters; initial substrate concentration, (100-200 g/L), temperature (28-32 °C) and pH (4.5-5.5). Initial substrate concentration, fermentation temperature and pH had a strong linear effect on levansucrase production. Regression model was significant (P0.05). Results of the statistical analysis showed that the fit of the model was good in all cases. Maximum levansucrase production of 13.3 mM glucose/min was obtained at the optimum levels of process variables (initial substrate concentration 159.01 g/L, fermentation temperature 30.3 °C and pH 4.91).
