Isolation, purification and identification of moderately halophilic bacteria and determination of some molecular properties

Bu çalışmada İzmir Çamaltı Tuzlası tuz yığını ve tuzlu su havuzundan su örneği ve İzmir Kuş Cenneti tuzlu su havuzundan su örneği ile ticari Himalaya tuz örneği gibi çeşitli tuz örneklerinden ılımlı halofilik ve halotolerant bakteriler izole edilmiş, yüksek tuzda yaşayabilen 10 izolat seçilerek saflaştırılmış, ayırtedici fenotipik ve biyokimyasal testleri ile 16S rRNA gen bölgesi çalışılmıştır. Sonuç olarak Gammaproteobacteria, Firmicutes, Actinobacteria şubelerine ait ve Halomonadaceae, Vibrionaceae, Bacillaceae, Micrococcaceae ve Enterobacteriaceae ailelerine ait suşlar elde edilmiştir. Bacillus sp. CT-2 suşu; Bacillus sp. CT-3 suşu; Bacillus sp. CT-4 suşu; Pantoea sp. CS-4 suşu; Oceanobacillus sp. HT-9 suşu; Salinivibrio sp. KC-2 suşu; Halomonas sp. KC-101 suşu ılımlı halofilik iken; Virgibacillus sp. CT-5 suşu, Kocuria sp. CT-6 suşu, Halomonas sp. CS-5 suşunun halotolerant karakterde olduğu tespit edilmiştir ve suşlar NCBI Gen Bankasına kaydedilmiştir. Tüm suşlar pH 5-11 arasında gelişen alkalitolerant özelliktedir. Suşların 9'u 4 ºC'de gelişen psikrotolerant özelliktedir. Yine suşların 9 tanesi 45, 50 veya 55 ºC'de gelişen termotolerant özelliklere sahiptir. Bu özellikleriyle ılımlı halofilik ve halotolerant suşların ülkemizde ilk defa olmak üzere çoklu ekstrem koşullara adapte olmuş Poliekstremotolerant bakteriler olduğu da ortaya konmaktadır. Kocuria sp. CT-6, Halomonas sp. CS-5 ve Oceanobacillus sp. HT-9 suşlarının ectBC gen bölgesi varlığı ile bu çalışmada dizayn edilen orijinal primerler kullanılarak tespit edilmiştir. Bu tez çalışması ile elde edilen suşların ectBC gen bölgesine sahip olduğu böylece potansiyel ektoin üreticisi oldukları ortaya konmuştur.In this study moderately halophilic and halotolerant were isolated from various salt samples such as Izmir Camalti Saltern salt bulk and salty water, Izmir Bird Paradise salty water and commercial Himalayan salt, the 10 isolates that can live at high salt concentrations were selected and purified. Differential phenotypical and biochemical tests were done and 16 S rRNA gene regions were analysed. Consequently, the strains were determined that they were in the phylums of Gammaproteobacteria, Firmicutes, Actinobacteria and in the families of Halomonadaceae, Vibrionaceae, Bacillaceae, Micrococcaceae ve Enterobacteriaceae. 7 of them; Bacillus sp. CT-2, Bacillus sp. CT-3, Bacillus sp. CT-4, Pantoea sp. CS-4, Oceanobacillus sp. HT-9, Salinivibrio sp. KC-2, Halomonas sp. KC-101 were detected as moderately halophilic and 3 of them; Virgibacillus sp. CT-5, Kocuria sp. CT-6, Halomonas sp. CS-5 were detected as halotolerant. The strains were submitted to NCBI GenBank. All of them were alkalitolerant characteristics in the ability of living at the range of pH 5-11. 9 of them were psychrotolerant characteristics in the ability of living at 4 ºC and 9 of them were thermotolerant characteristics in the ability of living at 45, 50 or 55 ºC. In addition to their moderately halophilic and halotolerant properties of these bacteria, this is the first report of Polyextremotolerant bacteria from Turkey which can tolerate multiple extreme conditions. Kocuria sp. CT-6, Halomonas sp. CS-5 and Oceanobacillus sp. HT-9 strains were positive for ectBC gene region analyses by using original primers designed in this study. According to the results these strains were detected as potential ectoine producers for further studies
