The effects of nigella sativa L. (black seed) oil on ovarian morphology and oxidant system in experimental diabetes

Bu çalışmanın amacı diyabetik ratlarda Nigella sativa L. yağının ovaryum hacmi, nuclear factor kappa-b (NF-κB) ve X-linked ınhibitor of apoptosis protein (XIAP) ekspresyonu ve serum malondialdehit (MDA), süperoksit dismutaz (SOD), total antioksidan durum (TAS) ve total oksidan durum (TOS) seviyesi üzerine etkisini araştırmaktır. Yirmi bir olgun dişi rat 3 gruba ayrıldı: kontrol grubu, diyabet grubu ve diyabet + Nigella sativa L. yağı grubu 0,2 mg/kg/gün Nigella sativa L. yağı haftanın 6 günü 30 gün boyunca verildi. NF-κB ve XIAP ekspresyonunu değerlendirmek için ovaryan kesitler immünohistokimyasal olarak analiz edildi. Ayrıca sağ ve sol ovaryan hacimleri steorolojik teknikler ile hesaplandı. Son olarak serum MDA, SOD, TAS, ve TOS seviyeleri saptandı. Sonuçlar Nigella sativa L. yağının hiperglisemiyi kontrol seviyesi kadar olmasa da düşürdüğü ortaya çıkardı. Nigella sativa L. yağının kontrol seviyelerine göre serum TOS ve MDA değerlerini düşürdüğü ve SOD ve TAS seviyelerini artırdığı gösterildiği için antioksidan özellikler sergilemiştir. Muhtemelen 30 günlük kısa diyabet süresine bağlı olarak guplar arasında total ovaryan hacimde ve XIAP ve NF-κB ekspresyonunda ciddi fark yoktu. Bu sonuçlar diyabetik hastalarda Nigella sativa L. yağının kan glukoz seviyesini ve oksidan aktiviteyi düşürmek için kullanılabileceğini akla getirmektedir.The aim of this study was to investigate the effects of Nigella sativa L. oil on ovary volume, NF-κB (Nuclear factor-kappa B) and XIAP (X-linked inhibitor of apoptosis protein) expression, and serum MDA (Malondialdehyde), SOD (Superoxide dismutase) TAS (Total antioxidant status) and TOS (Total oxidant status) levels in diabetic rats. Twenty-one adult female rats were divided into three groups: the control group, the diabetes group, and the diabetes + Nigella sativa L. oil group, which received 0.2 mg/kg/day Nigella sativa L. oil 6 days per week for 30 days. Ovarian sections were then analyzed using immunohistochemistry to assess NF-κB and XIAP expression. In addition, the right and left ovarian volumes were calculated using stereological techniques. Finally, serum MDA, SOD, TAS, and TOS levels were determined. The results revealed that Nigella sativa L. oil reduced hyperglycemia, but not to control levels. Nigella sativa L. oil also exhibited antioxidant properties, as demonstrated by lowered serum TOS and MDA values and increased SOD and TAS levels compared with the control. There were no significant differences in total ovarian volume or XIAP and NF-κB expression among the groups, possibly due to the short 30-day diabetes duration. These results suggest that Nigella sativa L. oil could be used to reduce blood glucose levels and oxidant activity in diabetic patients
