Effects of pregnant mare serum gonadotrophin (pmsg) and human chorionic gonadotrophin (hcg) induced follicular growth on vaginal smear findings in wistar rats

Bu çalışmada 25 erişkin dişi Wistar ratta siklus dönemlerine bakılmaksızın farklı doz ve aralıklarla gebe kısrak serum gonadotropini (PMSG) ve İnsan korionik gonadotropini (HCG) ile süperfollikülasyon ve süperovulasyon gerçekleştirildi. Ratlar dört deneme ve bir kontrol grubu olmak üzere beş gruba ayrılarak 0. saatten başlayarak sırasıyla I., II., III., IV. Gruplar ve Kontrol grubuna şu uygulamalar yapıldı: Grup I: 40 IU PMSG, 24. saatte 20 IU HCG; Grup II: 20 IU PMSG, 24. saatte 20 IU HCG; Grup III: 40 IU PMSG, 48. saatte 20 IU HCG; Grup IV: 20 IU PMSG, 48. saatte 20 IU HCG; Kontrol: Yalnız 4 IU PMSG. Bütün hayvanlarda 0. saatten başlayarak 12 saat ara ile 72. saate kadar vaginal smear örnekleri alınarak seksüel siklusların seyri ve vagina epitelinin egzojen gonadotropinlere duyarlılığı incelendi. Alınan smearlarda bazal/parabazal, intermedier, süperfisiyel ve keratinize süperfisiyel hücreler sayılarak yüzdeleri belirlendi. Çalışmada süperovulasyon dozlarında uygulanan PMSG’nin neden olduğu hiperöstrojenik etkinin vagina epitelinde, normal östrusta beklenenden daha hızlı değişimlere yol açtığı ve özellikle proöstrus aşamasının kontrol grubunda olduğundan daha hızlı şekillendiği görüldü. Ratlarda ovulasyonun göstergesi olarak sayılan kornifiye hücrelerin sayısındaki artışın PMSG ve HCG uygulamalarıyla uyum içerisinde şekillenmediği, bunun yerine tüm smearlarda 12 ila 60 saatlik bir döneme yayıldığı görüldü. Bu olgunun PMSG’nin uzun serum yarılanma ömrü nedeniyle şekillendiği kanısına varıldı.In this study Pregnant Mare Serum Gonadotrophin (PMSG) and Human Chorionic Gonadotrophin (HCG) were used to induce superfolliculation and superovulation on 25 adult Wistar rats without regarding the stage of sexual cycles. Rats were divided to a total of five groups as four trial and one control group and the following injections were given beginning on hour 0: Group I: 20 IU HCG 24 h. after 40 IU PMSG; Group II: 20 IU HCG 24 h. after 20 IU PMSG; Group III: 20 IU HCG 48 h. after 40 IU PMSG; Group IV: 20 IU HCG 48 h. after 20 IU PMSG; Control: Only 4 IU PMSG. Vaginal smears were taken with an interval of 12 hours, between 0 – 72 hours, from all of the animals in order to determine the progression of sexual cycle and the sensitivity of vaginal epithelium to exogenous gonadotrophins. The percentages of basal/parabasal, intermediate, superficial and cornified cells were determined by counting. It was observed that superovulatory doses of PMSG caused a more rapid progression than seen in normal cycles and the initiation of proestrus stage were faster than observed in the control group, owing to the hyperstrogenic effect on vaginal epithelium. It was observed that the rise in the percentage of cornified cells which is an indicator of ovulation were not in accordence with the injections of PMSG and HCG on the contrary it was distributed within a period of 12 to 60 hours. It was concluded that this effect was caused as a result of long serum half-life of PMSG.Intervet Veteriner İlaçları A.
