Histological investigation on the effects of feeding with a diet containing red hot pepper on m. Iliofibularis and M. Pektoralis of isobrown chicks

Çalışmada M. Pektoralis ve M. İliofibulariste kırmızı acı biberli rasyon ile beslenme nedeniyle glikojen ve lipid metabolizmasına bağlı olarak ortaya çıkabilecek olası histolojik değişiklikleri incelemek amaçlandı. Araştırma yumurtacı karakterde 40 adet Isobrown ırkı civciv ile başladı ve 5 ay devam etti. Kontrol grubu yumurtacı civciv gelişimine uygun rasyon ile beslendi. Deneme grubu, kontrol grubu rasyonuna %1 oranında Kahraman Maraş yöresi kırmızı acı biberi ilave edilerek beslendi. M. İliofibularis ve M. Pektoralisden alınan doku örnekleri Formol ve Bouin solusyonlarında tepit edildi. Parafin kesitlere yapısal özelliklerin tespit edilebilmesi için Mallory’nin Crossmonn tarafından modifiye edilen üçlü boyama, nötr mukopolisakkaritler için PAS, dondurma kesitlerine lipidler için Sudan Black B teknikleri uygulandı. Kontrol ve deneme gruplarında karkasın rengi bakımından makroskobik olarak bir farklılık görülmedi. Karın içi yağ miktarının özellikle 5. ay deneme gruplarında kontrollere göre az olduğu tespit edildi. Sudan Black B tekniği uygulanan dondurma kesitlerinde; tüm kontrol gruplarında M. İliofibularisin çoğunlukla lipidden zengin, M. Pektoralisin ise lipidden fakir kas tellerinden oluştuğu görüldü. Deneme gruplarında ise lipid miktarının çok az olduğu tespit edildi. Periodic Acide Schiff tekniği uygulanan preperatlarda kontrol ve deneme grupları arasında bir farklılık gözlenmedi. Sonuç olarak; rasyona %1 oranında Kahraman Maraş yöresi kırmızı acı biberi ilave edilmesi M. İliofibularis ve M. Pektoralisin lipid birikimini olumsuz yönde etkilerken, PAS reaksiyonu üzerinde bir etkisi olmadığı görüldü.This study was made on 40 Isobrown chicks and carried out for 5 months. The control group chicks were fed with the diets prepared according to the layers. The experimental group chicks were fed with the control group diet supplemented with 1% red hot pepper of Kahraman Maraş region. The tissue samples taken out from M. İliofibularis and M. Pektoralis were fixed in Bouin and Formalin solutions. Paraffin sections were stained with Crossmonn’s triple stain for the structural features, PAS stain neutral mucopolysaccarides and cryostat sections were stained with Sudan Black B method for lipids. Any macroscopical differences weren’t seen on the carcass colour in both the control and experimental groups. Abdominal fat content of the experimental group was less than that of the control group especially in the fifth month. In the cryostat sections, all of the control groups of were stained dense with Sudan Black B in M. Iliofibularis whereas it vicaverca in M. Pektoralis. The quantity of lipid was less in experimental groups. Any differences weren’t seen between the control and experimental group muscle sections applied with PAS stain. As a result; less lipid was seen in the experimental group chicks fed with a diet supplemented with 1% red hot pepper of Kahraman Maraş region
