This thesis describes efforts in the development of an immobilised enzyme biocatalyst for a scaleable biotransformation. The enzyme transketolase, which catalyses the synthesis of carbon-carbon bonds, was chosen as a model. The enzyme has been cloned into E.coli and can be produced on a large scale and is consequently representative of an industrial enzyme. Transketolase has been immobilised onto the commercially available materials Eupergit-C and Amberlite XAD7. With a binding yield of ca 90 % and a retention of activity between 20 % and 60 % depending upon enzyme load in the case of Eupergit-C and a binding yield of 90 % and a retention of 20% activity in the case of Amberlite XAD7.
