The effects of pollen, propolis and caffeic acid phenethyl ester on tyrosine hydroxylase activities and total rna levels in hypertensive rats caused by nitricoxide synthase blockage

Nitrik oksit (NO), enzimatik olarak L-Arjinin aminoasitinden sentezlenir. L-Arjinin analoglarından olan Nω-nitro-L-arginine methyl ester (L-NAME), Nitrik oksit sentaz (NOS) enzimini inhibe ederek organizmada NO üretimini azaltır ve bu durum hipertansiyona sebep olur. Katekolaminler stres durumunda salgılanan nörotransmitterlerdir. Katekolaminlerin salgılanmasında ilk basamak tirozin aminoasitinin tirozin hidroksilaz enzimi aracılığıyla L-3,4 dihidroksifenilalenine (LDOPA) dönüştürülmesidir. Stres durumuna tirozin hidroksilaz (TH) enzim aktivitesinde artış görülmektedir. Propolis, polen ve kafeik asit fenetil ester (CAPE) fenolik bileşikler açısından zengin arı ürünleridir. Bu çalışmanın amacı L-NAME uygulaması ile NOS inhibisyonu gerçekleştirilerek geliştirilen kronik hipertansif Sprague Dawley türü erkek sıçanlara arı ürünlerinin uygulanması sonucu bu ürünlerin sıçanların adrenal medulla, hipotalamus ve kalp dokularında TH enzim aktivitesi, total RNA miktarı ve total protein miktarı üzerine etkilerini incelemektir. Deney sonuçları gösteriyor ki LNAME uygulaması ile TH enzim aktivitesi artmaktadır ve arı ürünlerinin uygulanması sonucu TH enzim aktivitesi azalmaktadır.NO is enzymatically synthesized from L-arginine, a process that can be antagonized by subrogated L-arginine compounds such as Nω-nitro-L-arginine methyl ester (LNAME), which compete for the NO synthase (NOS). Inhibition of NOS reduces NO production and this inhibition caused vasoconstraction. Catecholamines are neurotransmitters which are releasing in stress conditions. Tyrosin hydroxylase has a key role in catecholamine biosyntesis. This enzyme catalyzing a reaction which is converting tyrosine aminoacid to L-3,4-dihydroxyphenylalanine (L-DOPA). Stress effects TH enzyme activity. stress conditions increased TH activity. Propolis, pollen and CAPE are natural bee products and this products are rich in polyphenols. The objective of the present study was to evaluate the effects of propolis, pollen, and caffeic acid phenethyl ester (CAPE) on tyrosine hydroxylase (TH) activity, total protein levels and total RNA levels of Nω-nitro-L-arginine methyl ester (L-NAME) inhibition of nitric oxide synthase in the heart, adrenal medulla, and hypothalamus of hypertensive male Sprague Dawley rats. All this datas shows that L-NAME treatment increases TH enzyme activity and natural bee products such as propolis, pollen and CAPE modulates the TH enzyme activity
