Screening of some Streptomyces species in view of lipase inhibitors and optimization of fermentation media

Bu çalışmada, bazı Streptomyces suşlarının üretmiş olduğu sekonder metabolitlerin, domuz pankreatik lipazına karşı gösterdiği inhibitör etki incelendi. Bu amaçla Streptomyces griseus subsp. griseus DSM41107, S. toxytricini DSM40178, S. griseus subsp. griseus DSM40693, S. rimosus subsp. paramomycinus DSM41429, S. auratus DSM41897, S. fradiae DSM40063, S. antibiticus DSM40234 suşları Alman Kültür Koleksiyonu?ndan (DSMZ) temin edildi. Mikroorganizmalarının metaboliti üretmesi için optimum şartlar sağlandı. Streptomyces griseus subsp. griseus DSM41107 suşunun M6 besiyerinde üretmiş olduğu biyomolekülün lipaz inhibisyonu sağladığı tespit edildi. Fermentasyon sıvısı etil asetat ile ekstrakte edildi. Ardından inhibisyonda etkili olan metabolit(ler) kolon kromotografisi ile kısmi olarak saflaştırıldı. HPLC?de yaklaşık 12. dakikada toplanan fraksiyon orlistat ile karşılaştırmalı olarak test edildi. Test sonucunda, fraksiyonun 5 mg/mL konsantrasyonu Domuz Pankreatik Ekstraktını % 90 oranında, aynı konsantrasyonlu orlistatın ise % 99.92 oranında inhibe ettiği saptandı. Ayrıca; aynı fraksiyonun 7 mg/mL konsantrasyonunun C. rugosa ve M. javanicus lipazlarını sırasıyla % 40 ve % 59 oranında inhibe ettiği tespit edildi. In this study, secondary metabolites produced by certain strain of Streptomyces genus analyzed inhibitory effects against porcine pancreatic lipase. For this purpose, Streptomyces griseus subsp. griseus DSM41107, S. toxytricini DSM40178, S. griseus subsp. griseus DSM40693, S. rimosus subsp. paramomycinus DSM41429, S. auratus DSM41897, S. fradiae DSM40063, S. antibiticus DSM40234 were obtained from German Culture Collection (DSMZ). It was provided optimum conditions for the microorganisms to produce the metabolite. It was detected lipase inihibiton by the biomolecule(s) that was produced in M6 medium by Streptomyces griseus subsp. griseus DSM41107. Fermentation broth was extracted by ethyl acetate. Then the metabolite(s) that effective on inhibition were partially purified by coloumn chromatography. Fraction that collected about 12th second at HPLC was tested in comparison with orlistat. As a result of the test, it was determined to be inhibited porcine pancreatic lipase by the consantration of 5 mg/mL extract at rate of 90%, in case of the same consantration of orlistat was 99.92%. Also the 7 mg/mL consantration of the same fraction was detected to inhibited at rate of 40% and 59% C. rugosa and M. javanicus lipases, respectively.
