We have used an animal model of depression, learned helplessness; to attempt to define the circuit mediating helplessness, determine the cellular basis underlying these circuit changes and the pathology leading to these changes. Using the learned helpless model two rat lines were created by selectively breeding rats either sensitive or resistant to helplessness training selectively with one another. The helpless line has the characteristics of a treatment resistant depression not responding to normal antidepressants or ECT, whereas the non helpless line is resistant to the effects of uncontrollable stress.
