Research concerning the feasibility of replacing antibodies with binding proteins as the specific binders in enzyme-linked competitive binding assays is described. The relative merits of these techniques compared to traditional enzyme immunoassays are presented for both heterogeneous and homogeneous arrangements. The folate binding protein (FBP)-folate system is used as a model to demonstrate the analytical properties of such systems. A theoretical model for heterogeneous enzyme-linked assays is detailed. This model compares the use of antibodies and binding proteins as binders in these assays and demonstrates that by using such binding reagents the overall dose-response characteristics of the assays could be improved, particularly when ...
