Development of Magnetic Resonance Imaging tools for the study of the dog, horse and sheep in neuroimaging

De plus en plus fréquemment, les études précliniques sur modèle animal utilisent les acquisitions IRM. Cependant, l'acquisition d'images à partir d'un cerveau animal présente certaines différences avec celle réalisée chez l'Homme, qui demande donc des adaptations des séquences. Une fois l'acquisition réalisée, le second problème concerne les outils et la méthodologie disponible pour le post-traitement des images. Effectivement, ces outils existent tous chez l'Homme (Template, Atlas, Outils de normalisation et segmentation), mais peu de modèles animaux en disposent. Ainsi, la littérature et les ressources méthodologiques sont très pauvres en ce qui concerne le Chien, pourtant de plus en plus utilisé comme modèle d'étude en neurosciences. Dans cette étude, nous nous sommes principalement concentrés sur la mise en place de ces outils chez le Beagle. Pour cela, 23 chiens de laboratoire sans troubles neurologiques ont été anesthésiés pour passer des acquisitions dans une IRM 3 Tesla. Le protocole d'acquisition consistait en une pondération T1 et T2, et DTI. Pour la première fois, des Templates pondérés T1, T2 et DTI à la fois in-vivo et post-mortem ont été mis en place. De plus les Templates DTI permettent une tractographie et ainsi plusieurs ensembles de fibres ont pu être reconstruits (Cingulum, Corpus Callosum, Fornix, Capsule Interne et Commissure rostrale). En plus des indices IRM, des coupes histologiques ont été réalisées à partir d'un encéphale de Beagle provenant de la cohorte d'étude. Deux colorations ont été réalisées : une pour la myéline et l'autre pour la substance de Nissl. Les coupes histologiques ont été corrélées avec les différents Templates IRM. Le principe est de permettre à un utilisateur d'avoir la correspondance d'un voxel du Template ou d'une de ces acquisitions normalisées avec les coupes histologiques. De plus, cette étude a aussi permis la mise en place de différents outils méthodologiques qui ont pu être adaptés à l'étude d'autres espèces animales : le Chat, la Brebis et le Cheval.More and more frequently, preclinical studies on animal models use MRI acquisitions. However, the acquisition of images from an animal brain has some differences with that conducted in humans, which therefore requires adaptations of the sequences. Once the acquisition is completed, the second problem concerns the tools and methodology available for image post-processing. In this study, we mainly focused on the implementation of these tools in the Beagle. For this, 23 laboratory dogs without neurological disorders were anesthetized to make acquisitions in a 3 Tesla MRI. The acquisition protocol consisted of T1 and T2 weighting, and DTI. For the first time, T1, T2 and DTI weighted templates both in-vivo and post-mortem have been implemented. In addition, the DTI Templates allow a tractography and thus several sets of fibers could be reconstructed (Cingulum, Corpus Callosum, Fornix, Internal Capsule and Rostral Commissure). In addition to the MRI indices, histological sections were taken from a Beagle brain from the study cohort. Two stains were performed: one for myelin and the other for Nissl's substance. The histological sections were correlated with the different MRI Templates. The principle is to allow a user to have the correspondence of a Template voxel or one of these normalized acquisitions with histological sections. In addition, this study has also enabled the implementation of various methodological tools that have been adapted to the study of other animal species: Cat, Sheep and Horse
