Dna extraction technique from the silk gland of the silk worm bombyx morí (lepidoptera: bombycidae)

Selection for production of puré lines of the silkworm (Bombyx morí), starting with commercial hybrids, is a long and expensive process, however, molecular biology techniques can shorten the time and allow to determine favorable characteristics for silk production. The objective of this work consisted in establishing a protocol for the extraction of DNA of good quality and quantity of the silkworm. The research was carried out at the molecular biology laboratory of the Ü.D.C.A at Bogotá, using larvae coming from the breeds of segregant lines maintained in the laboratory. For the extraction of DMA the posterior, médium and anterior part, placed in a Buffer extraction solution with Tris HCI, MaCI and EDTA pH 8, of the silk gland was used. Subsequently the samples were washed with phenol chloroform and isoamylic chloroform. For the DMA precipitation frozen ethanol was applied. Electrophoresis in agarosa was performed and quantified in a mini-fluorometer with a fixed longitude wave, obtaining a range between 0.21/xg/ml and 1.5/xg/ml of DMA. The quality and quantity of the DMA of the posterior part of the silk gland of five day oíd larvae of the last instar was superior to that obtained from larvae of other ages and other segments of the gland. Therefore the posterior part of the silk gland is considered to be the most adequate tissue for the DMA extraction process related to molecular analysis of silk productivity components.La selección a partir de híbridos comerciales para la producción de líneas puras del gusano de seda (Bombyx morí) es un proceso largo y costoso, sin embargo, las técnicas de la biología molecular acortan el tiempo y permiten determinar las características favorables para la producción de seda. El objetivo de este trabajo consistió en establecer un protocolo para la extracción de ADN de buena calidad y cantidad. La investigación, se realizó en el laboratorio de Biología Molecular de la Ü.D.C.A, en Bogotá, utilizando larvas provenientes de las crías de líneas de segregantes mantenidas en el laboratorio. Para la extracción del ADN, se utilizó la parte posterior, media y anterior de la glándula de seda, la cual se colocó en una solución Buffer de extracción con Tris HCI, NaCI y EDTA pH 8; posteriormente, las muestras se sometieron a lavados con fenol cloroformo y cloroformo ¡soamílico. Para la precipitación del ADN, se aplicó etanol congelado. Se corrió la electroforesis en agarosa y se cuantificó en el mini-fluorómetro con una longitud de onda fija obteniéndose un rango entre 0,21/¿g/ml y 1,51 /¿g/ml de ADN. La cantidad y calidad del ADN encontrado en la parte posterior de la glándula de seda en larvas de cinco días del último instar larval fue superior a la que se encontró en larvas de otras edades y en diferentes segmentos de la glándula, por lo cual, es un tejido adecuado para los procesos de extracción de ADN relacionados con el análisis molecular de componentes de productividad.Incluye referencias bibliográfica
