Potential of bacterial isolated from the roots of fig and pirobetuminoso shales in the control of Meloidogyne incognita in Ficus carica cv. Roxo de Valinhos

The fig tree (Ficus carica L.) is a fruitfull grown throughout the world due to its ability to climate adaptation; being important economically to Brazil and Rio Grande do Sul State in the family farming context. However, pests such as the root-knot nematode Meloidogyne incognita seriously affect the fruit production considering the high aggressiveness of this nematode and the lack of effective management technologies for control of this pest. In this context, the phytosanitary management in infested fig orchards by this nematode is essential to allow the economic viability for this fruit crop. Thereby, the introduction of biological control in the M. incognita integrated management is an important strategy. Among biocontrol agents, the rhizobacteria are good rhizosphere competent bacteria for promoting plant growth and or inhibiting the action of parasitic nematodes on host plants. Therefore, the objectives of this study were: 1) To isolate bacteria with potential for biocontrol of M. incognita from the fig tree roots and pirobetuminoso shale rocks; 2) to characterize the most promising isolates through biochemical tests and DNA sequencing; 3) to evaluate the performance of selected bacterial isolates in the biocontrol of M. incognita on fig tree growing in naturally infested soil under two application forms of the microorganisms; and 4) to evaluate the effect of treatments on bacterial inhabitant nematofauna soil. Firstly, the bacterial isolates obtained from the pirobetuminoso shales and from the fig roots were evaluated on the hatching and mortality of M. incognita second-stage juveniles in in vitro tests besides the production of related compounds to biological control of nematodes. Fourteen bacterial isolates produced at least one compound related to the biocontrol of plant parasitic nematodes (protease Litmus milk and gelatin, lipase, chitinase and or ammonia), and high levels of mortality (78-100 %) or inhibition of hatching (73-100 %) of second stage juveniles of the nematode. Subsequently, these bacterial isolates were selected, characterized via biochemical tests and DNA sequencing, and then they were evaluated for their potential for the RKN biocontrol and fig growth promotion in two experiments carry out in the greenhouse at different application forms of antagonists. In the first experiment, root systems of 'Roxo de Valinhos' nursery fig plants were microbiolized with 14 bacterial, separately, and 30 days after, each fig plant was transplanted into pot with soil 8 naturally infested with M. incognita. Although the root-knot nematode reduced the development of the fig trees in all treatments, the application of the isolates F25 ( Bacillus sp.) , F78 (Streptomyces sp.) , F76 (Streptomyces sp.) , FB59 (Pseudomonas denitrificans) and FB34 (Arthrobacter oxydans ) declined the root-knot nematode reproduction (P < 0.05 ) rate in the fig plants at 49, 46 , 39 , 28 and 26 %, respectively, comparing to the plants not inoculated and non microbiolized. In the second experiment, two bacteria (F75 and F78) more promising as M. incognita potential biocontrollers in the in vitro tests, were evaluated for its capacity to suppress the RKN and promoting growth of the fig plants. For this purpose, 24 hours before planting the fig nurseries, pots containing soil naturally infested with M. incognita were sprayed with the respective bacterial suspensions of both isolates, separately. The bacterial isolates suppressed the reproduction of M. incognita at rate of 21 to 43% comparing to the control where the non microbiolized fig plants were kept in infested soil. However, additional researches in this pathosystem will enable a better understanding of action mechanisms of the tested bacteria on the fig tree and the insertion of this technique in a program of root-knot nematode integrated management.A figueira (Ficus carica L.) é uma frutífera cultivada em todo mundo devido a sua capacidade de adaptação climática; sendo essa economicamente importante para o Brasil e para o Rio Grande do Sul no contexto da agricultura familiar. No entanto, pragas como o nematoide das galhas (NG) Meloidogyne incognita afetam seriamente a produção considerando a elevada agressividade do parasita e a falta de tecnologias de manejo efetivas para o controle dessa praga. Nesse contexto, o manejo fitossanitário de pomares de figueira infestados por esse nematoide é indispensável para a cultura seja viável economicamente. Assim, a inserção do controle biológico no manejo integrado de M. incognita constitui-se como uma estratégia importante. Entre os biocontroladores, as rizobactérias são bactérias competentes da rizosfera por promover o crescimento vegetal e ou inibir a ação parasítica dos fitonematoides sobre as plantas hospedeiras. Dessa forma, os objetivos desse trabalho foram: 1) Isolar bactérias com potencial para o biocontrole de M. incognita a partir de raízes de figueira e de rochas de folhelhos pirobetuminosos; 2) Caracterizar os isolados mais promissores através de testes bioquímicos e sequenciamento de DNA; 3) Avaliar o desempenho dos isolados bacterianos selecionados, no biocontrole de M. incognita em figueira cultivada em solo naturalmente infestado, sob duas formas de aplicação; e, 4) Avaliar a influência dos tratamentos bacterianos na nematofauna habitante do solo. Inicialmente, os isolados obtidos de folhelhos pirobetuminosos e de raízes de figueira foram avaliados em ensaios in vitro quanto ao efeito sobre ovos e juvenis de segundo estágio de M. incognita, e, á produção de compostos relacionados ao controle biológico de nematoides. Quatorze bactérias produziram pelo menos um composto relacionado ao biocontrole de fitonematoides (protesease em gelatina e leite de Litmus, lípase, quitinase e ou amônia), e resultaram em altos níveis de mortalidade (78-100%) ou inibição da eclosão (73-100%) dos juvenis de segundo estádio do nematoide. Estes isolados foram selecionadas e caracterizadas através de provas bioquímicas e sequenciamento de DNA, sendo, a seguir, avaliadas quanto ao biocontrole do NG e promoção de crescimento de figueiras em dois ensaios conduzidos em casa de vegetação sob diferentes formas de aplicação dos antagonistas. No primeiro ensaio, raízes de mudas de figueira 'Roxo de Valinhos' foram microbiolizadas com as 14 bactérias selecionadas, separadamente; e, 30 dias 6 após, cada planta foi transplantadas individualmente para vasos com solo naturalmente infestado com M. incognita.
