Anti cancer effects of newly synthesized zno/Ag zno nano composites on human gastric cancer cell line (AGS) and melanoma cancer cell line (A-375)

Purpose This study accomplished to examine anti cancer effect of zno nanoparticles and Agzno nano composites on melanoma and gastric carcinomacancer cell line. Methods: Human gastric carcinoma (AGS) and human melanoma (A-375) cancer cell lines treated by different concentrations of Ag zno/zno nano composites cytotoxcity examined through MTT and colnogenic assays ethidium bromide/acridine orange (EB/AO) staining used for apoptotic cell detection. Adiamidino-2-phenylindole staining method was used to analysis cell cycle by flow cytometry. Results: The IC50 values, obtained for AGS and A375 and cell proliferation were assessed by MTT. Ethidium bromid/acridine orange staining demonstrated that different concentrations of zno nanoparticles and Ag/ zno nano- composites its cytotoxic effect via apoptosis. The mechanism for the inhibition was further investigated with cell cycle analysis, during analysis cell cycle by flow cytometry, it was found that zno/Agzno nano composites, in the different phases stops the cell cycle. Meanwhile, zno nanoparticles and Ag/ zno nano- composites are able to induce cytotoxic effect via cell cycle arrest at S and G1 phase. Conclusion: Thus, zno nanoparticles and Ag/ zno nano- composites were ability to induce cytotoxicity in the human melanoma (A-375) and human gastric carcinoma (AGS) cancer cell line in the lower micromolar concentrations. In conclusion, this finding may introduce a new view on the mode of action and possible application of new nano composites for cancer treatment
