In the yeast Saccharomyces cerevisiae the PEP4 gene product, protease A, is responsible for activating all soluble vacuolar (lysosomal) enzymes. These vacuolar enzymes remain inactive in pep4 mutants. Vacuolar trehalase activity was diminished in such mutants as well.
