Background: In this study we use molecular docking to find compounds that interact with the enzyme tryptophan hydroxylase (TPH), followed by experimental validation. TPH is a tetrahydrobiopterin (BH4) dependent enzyme that catalyze the conversion of L-tryptophan to 5-hydroxytryptophan (5-OH-Trp). The reaction is the first and also rate limiting step in the synthesis of serotonin (5-hydroxytryptamine, 5-HT) which is precursor for the biosynthesis of melatonin.
