3. SUMMARY Background: The origin and genesis of the autophagosomal membrane is still a matter of debate. There are indications that the phospholipid phosphatidylinositol 3-phosphate (PI3P) may play a pivotal role in early formative stages, by acting as a recruiter of secondary proteins containing PI3P binding FYVE domains (including DFCP1). This project seeks to investigate the role of PI3P in autophagy; where it is located, when it is needed, and how autophagy (as measured by readouts of the autophagosome marker protein LC3) can be regulated by varying levels of PI3P.
